摘要
目的:优选北虫草可溶性蛋白质的最佳提取工艺。方法:对北虫草蛋白质提取条件进行单因素考察,并采用L9(34)正交试验设计法及考马斯亮蓝染色法,以北虫草可溶性蛋白质含量为指标,考察p H值、提取温度、料液比、提取时间4个因素对可溶性蛋白质提取率的影响,从而确定北虫草蛋白质的最佳提取工艺。结果:建立了北虫草蛋白质的最佳提取工艺:向北虫草药材粉末中加入60倍量p H为10的蒸馏水,在温度30℃条件下回流提取70 min。结论:优化的提取工艺稳定、合理、可行,能正确反映北虫草药材中的蛋白质含量,为北虫草生物活性物质的进一步研究提供了实验依据。
Objective : To optimize the extraction technology of protein in Cordyceps Militaris. Methods : The optimized extraction technology was investigated by using single factor test and orthogonal test with the content of water - soluble protein as the index for the first time which was determined by the method of coomassie brilliant blue staining. In this study, the effects of pH, tempera- ture, solid -liquid ratio and time on the extraction rate were investigated. Results :The optimized extraction technique of protein in Cordyceps Militaris was established, which was pH = 10, solid- liquid ratio 1:60 and 70 rain for extraction at the temperature of 30℃. Conclusion:The optimal extraction condition was stable and reliable and could provide an experimental basis for further study of the bioactive substance of Cordyceps Militaris.
