CD147和MMP-9在胃癌组织中的表达及其与预后的关系

Expression of CD147 and MMP-9 in gastric carcinoma and the correlation to prognosis

目的:探讨细胞外基质金属蛋白酶诱导因子(extracellular matrix metalloproteinase inducer,CD147)和基质金属蛋白酶-9(matrix metalloproteinases-9,MMP-9)在胃癌中的表达,两者的关系及其与胃癌浸润、转移和预后的关系。方法:应用免疫组化SP法检测63例胃癌组织及40例癌旁正常组织CD147、MMP-9蛋白的表达,并与临床病理参数和预后进行对比。结果:胃癌组织中CD147和MMP-9的阳性表达率分别为77.8%和81.0%,均明显高于癌旁正常组织中的表达率(25.0%和25.0%)(P均<0.05)。CD147和MMP-9的表达都与肿瘤浸润深度、TNM分期、淋巴结转移及Ki-67增殖指数相关(P<0.05)。CD147和MMP-9的表达呈正相关(r=0.907,P=0.000)。单因素生存分析显示CD147和MMP-9表达阳性的胃癌患者生存率低于表达阴性组。结论:CD147和MMP-9与胃癌侵袭和转移有关,CD147与MMP-9的产生有密切的关系,两者可作为胃癌判断临床预后的参考指标。

Objective： To investigate the expression of extracellular matrix metalloproteinase inducer（ CD147） and matrix metalloproteinases-9（ MMP-9） in human gastric carcinoma,and to study their relationship with the invasion,metastasis and prognosis. Methods： CD147 and MMP-9 protein expressions were examined by immunohistochemistry in 63 cases of human gastric carcinoma and 40 cases of paracancerous normal tissues,and the results were compared to clinical pathological factors and prognosis. Results： The expression rates of CD147 and MMP-9 in gastric carcinoma tissues were 77. 8% and 81. 0%,which were significantly higher than those in paracancerous normal tissues（ 25. 0% and 25. 0%）（ P 0. 05）. The expressions of CD147 and MMP-9 were significantly correlated with the depth invasion,TNM stages,lymph node metastasis and Ki-67 expression（ P 0. 05）. CD147 expression was positively correlated with MMP-9 expression（ r = 0. 907,P = 0. 000）. Univariable survival analysis indicated that overall survival rate was lower in patients with positive expression of CD147 and MMP-9 than in patients with negative expression. Conclusion： CD147 and MMP-9 may play a role in invasion and metastasis of gastric carcinoma. Increased MMP-9 might be induced by CD147 expressed in gastric tumor cells. They may be prognostic factors of gastric carcinoma.
