摘要
【目的】分析地处中国南方的重庆地区中华蜜蜂Apis cerana cerana(简称"中蜂")囊状幼虫病病毒(sacbrood virus,SBV)(Ac SBV-S.Chn-CQ)的遗传特性。【方法】采用RT-PCR法对采自重庆23个区县的阳性中蜂幼虫进行SBV多聚蛋白基因扩增、序列测定和序列分析,采用邻接法(neighbor-joining method)基于基因组和基因组片段SB11-SB12构建系统进化树,使用RDP3检测SBV的基因重组。【结果】Ac SBV-S.Chn-CQ与中国南方中蜂SBV(Ac SBV-S.Chn)其他分离株、越南北部东方蜜蜂A.cerana SBV(Ac SBV-N.Vie)、越南北部西方蜜蜂A.mellifera SBV(Am SBV-N.Vie)、韩国东方蜜蜂SBV(Ac SBV-S.Kor)的同源性较高。Ac SBV-S.Chn-CQ在结构蛋白编码区域存在连续51个核苷酸缺失,在非结构蛋白编码区域有3个不连续的核苷酸缺失,与Ac SBV-N.Vie,Am SBV-N.Vie和Ac SBV-S.Kor核苷酸缺失相同。Ac SBV-S.Chn-CQ与Ac SBV-S.Chn-FZ,Ac SBV-N.Vie,Am SBV-N.Vie和Ac SBV-S.Kor在亚洲基因型中形成一亚型,亚型内检测到重组事件。【结论】推测Ac SBV-S.Chn-CQ与SBV-N.Vie和Ac SBVS.Kor可能来自一个共同祖先。
【Aim】This study aims to analyze the genetic characteristics of sacbrooe virus( SBV) isolated from Apis cerana cerana in Chongqing,southwestern China( Ac SBV-S. Chn-CQ). 【Methods 】 The polyprotein gene of SBV was amplified by RT-PCR from samples of A. c. cerana larvae infected with SBV,which originated from 23 districts or counties in Chongqing,sequenced and analyzed. Phylogenetic trees were constructed using neighbor-joining method based on SBV genome,and the genomic region SB11-SB12. RDP3 was used to detect the genetic recombination. 【Results】Ac SBV-S. Chn-CQ showed high homologies with other SBV isolates from A. c. cerana in South China( Ac SBV-S. Chn),the SBV infecting A. cerana and A. mellifera in North Vietnam( Ac SBV-N. Vie and Am SBV-N. Vie) and the SBV infecting A. cerana in South Korea( Ac SBV-S. Kor). Ac SBV-S. Chn-CQ has a consecutive 51-nucleotide deletion along the structural protein-coding region and an inconsecutive 3-nucleotide deletion along the nonstructural protein-coding region,and its nucleotide deletion is identical to those of Ac SBV-N. Vie,Am SBV-N. Vie and Ac SBV-S. Kor. Ac SBV-S. Chn-CQ,Ac SBV-S. Chn-FZ,Ac SBV-N. Vie,Am SBV-N.Vie and Ac SBV-S. Kor formed a subtype within the Asian genotype,and recombination events were detected in the isolates within this subtype. 【Conclusion】Ac SBV-S. Chn-CQ,SBV-N. Vie and Ac SBVS. Kor may share a common origin.
