蟛蜞菊内酯大鼠血浆蛋白结合率的测定

Determination of the Binding Rate of Rat Plasma Protein with Wedelolactone

目的:测定蟛蜞菊内酯与大鼠血浆蛋白的结合率。方法:采用平衡透析法模拟蟛蜞菊内酯与大鼠血浆蛋白的结合过程,建立HPLC-UV法测定蟛蜞菊内酯与大鼠血浆蛋白的结合率。结果:在透析24 h后,蟛蜞菊内酯在低(0.62μg·m L-1)、中(3.10μg·m L-1)、高(9.92μg·m L-1)3个质量浓度下与大鼠血浆蛋白的结合率分别为(87.9±2.1)%、(84.7±2.5)%、(83.8±1.9)%。结论:蟛蜞菊内酯与大鼠血浆蛋白有较强的结合作用。

Objective: To study the binding rate of plasma protein with wedelolactone, an accurate and specific high performance liquid chromatography method (HPLC) for quantifying wedelolactone in rat plasma was established. Methods: The equilibrium dialysis combined with HPLC was carried out for the determi-nation of the plasma protein binding rate of wedelocatone. Results: 24 h after equilibrium dialysis, the binding rates of plasma protein with wedelolactone at low (0.62μg·mL-1), middle (3.10μg·mL-1) and high (9.92μg·mL-1) concentration were (87.9±2.1)%, (84.7±2.5)% and (83.8±1.9)%, respectively. Conclusion:These results showed the strong binding power of wedelolactone with rat plasma protein.
