摘要
为研究光照周期对黄鳝不同组织的基因组DNA甲基化的影响,设置3个不同的光照时间(8、10和12h)处理,对黄鳝心、肝、脾、肾、肠、肌肉和脑等组织基因组DNA甲基化用甲基化敏感扩增多态性(methylation sensitive amplified polymorphism,MSAP)技术进行分析。结果显示,不同组织基因组DNA对光照时间有不同的反应。其中,肌肉基因组DNA甲基化对光照周期的变化较为敏感,不同光照周期下肌肉基因组DNA总甲基化差异显著,光照周期为12h半甲基化和其他处理相比差异也很显著;肾脏和脑的基因组DNA甲基化对光照周期变化最不敏感,各处理组之间全甲基化、半甲基化和总甲基化差异都不显著;肝脏、肠、脾和心脏表现比较复杂,其中肝脏不同处理下基因组DNA半甲基化和总甲基化差异都不显著,光照周期为8h时,全甲基化差异显著;肠不同处理下基因组DNA全甲基化、半甲基化差异不显著,在光照周期为10h的处理组的总甲基化和其他处理组差异显著;脾脏在光照周期为10h和8h时全甲基化和总甲基化各处理组差异显著,在光照周期为12h时半甲基化和其他处理组差异显著;心脏在光照周期为10h时半甲基化和其他处理组差异显著。
To study the effect of light application time on genomic DNA methylation in rice field eel(Monopterus albus),three light application time(8h,10 h,12h)were arranged and the genomic DNA methylation patterns of seven tissues(heart,liver,muscle,brain,spleen and kidney)were analyzed by MSAP(methylation sensitive amplified polymorphism)technology.The results showed that different tissues had different methylation patterns among three treatments.The muscle was sensitive to light application time,in which total DNA methylation was significant different among three treatments and hemimethylation of 12 htreatment was significant different.Both the Kidney and brain were not sensitive to light application time,with no significant differences found in hemimethylation,methylation and total methylation among three treatments.Complicated changes were found in the liver,intestine,spleen and heart.In the liver,no significant differences of hemimethylation and total methylation were found among three treatments,while methylation of 8hwas significant different from the other two treatments.In the intestine,no significant differences of hemimethylation and methylation were found among three treatments while significant change of total methylation was found between 10 hand the other two treatments.In the spleen,significant differences of hemimethylation and total methylation were found between 10 hand 8htreatments,while hemimethylation of 12 hwas different from the other two treatments.In the heart,significant differences of hemimethylation were found in the treatment of 10 h.
