园林植物报春花的组织培养

Tissue Culture of Primula malacoides Franch.

本试验以报春花叶片、茎段作为外植体,进行了初代组织培养,并进行增殖、生根培养。结果表明:75%的酒精(0.5 min)+0.1%升汞(8 min)具有很好的消毒作用;MS+6-BA 1.0 mg·L-1+NAA 0.2 mg·L-1是不定芽分化的最佳培养基配比;MS+6-BA 1.0mg·L-1+NAA 0.2 mg·L-1是不定芽增殖的最佳培养基配比;1/2MS+NAA 0.2 mg·L-1是生根的最佳培养基配比。

In this experiment, leaves and stems of Primula malacoides Franch. were taken as explants, and conducted the primary tissue culture, and proliferation, rooting culture. The results showed that 75% alcohol（0.5 min） 0.1% mercuric chloride（8 min） had good disinfection effect; the best medium ratio for the differentiation of adventitious buds was MS ＋6-BA 1.0 mg·L-1＋NAA 0.2mg·L-1; for adventitious bud proliferation, the optimum culture medium composition was MS＋6-BA 1.0 mg·L-1＋NAA 0.2 mg·L-1;1/2MS＋NAA 0.2 mg·L-1was the best rooting culture medium composition.
