摘要
目的本研究通过检测比较microRNA-130b-5p(miR-130b-5p)在肾癌及癌旁正常组织之间以及肾癌细胞系和正常肾细胞系之间的表达水平,并分析其表达水平与临床病理资料之间的关系,为进一步研究miR-130b-5p在肾癌中深入的功能和机制奠定基础。方法采用qPCR方法检测miR-130b-5p在42对肾癌组织和癌旁正常组织中表达量的差异,并运用秩和检验方法分析肾癌组织中miR-130b-5p表达量与患者临床病理特征之间的相关性。培养肾癌细胞系(786-O和ACHN)和正常肾上皮细胞系(293T)并采用qPCR方法检测细胞中miR-130b-5p的表达量。结果miR-130b-5p在肾癌组织中的表达量明显高于癌旁正常组织(P<0.01),且肾癌组织中miR-130b-5p表达量与患者AJCC临床分期呈正相关(P=0.041),与其他临床病理资料如年龄、性别、T分期、组织类型和Fuhrman分级未发现明显相关关系(P>0.05)。同时,miR-130b-5p表达量在肾癌细胞系中也明显高于正常肾上皮细胞系(P<0.01)。结论 miR-130b-5p在肾癌组织和细胞系中均高表达,且其在肾癌组织中的表达水平与患者AJCC临床分期呈正相关,提示miR-130b-5p与肾癌的发生、发展相关,有望成为新的肾癌肿瘤标志物。
Objective To explore the function and mechanism of miR-130b-5p in renal cancer, we compared the expression level of microRNA-130b-Sp (miR-130b-Sp) between renal cancer and adjacent normal kidney tissues (ANTs) and between cells derived from renal cancer and normal kidney and we also analyzed the relationship between the expression level of miR-130b-5p and the clinical pathology. Methods Utilizing quantitative real-time polymerase chain reaction (qPCR) we determined the expression of miR- 130b-5p in 42 paired renal cancer tissues and ANTs and we analysed the relationship between miR-130b-Sp expression and clinical pathology by rank sum test. Cultivated the renal cancer cell lines (786-0 and ACHN) and human normal kidney cell line HK-2 and determined the miR-130b-5p expression level of cells by qPCR. Results MiR-130b-5p was up-regulated in RCC tissues and cell lines compared with control normal tissues and cell line. And the miR-130b-Sp expression in RCC tissues is correlated with AJCC stage (P =0.041), but there is no correlation was found between miR-130b-Sp expression and other clinical pathological variables such as age, sex, T stage, histological type and Furman grade (P〉0.05). Conclusions MiR-130b-5p was up-regulated in both renal cancer tissues and cell lines and miR-130b-Sp expression level was correlated to AJCC stage of renal cancer. It is suggested that miR-130b-5p plays a potential role in the initiation and development and miR-130b-Sp might be a no- vel biomarker in renal cancer.
