miR-10b在肾癌中的表达及临床意义研究

摘要 目的检测35例肾癌标本及配对正常组织标本中miR-10b的表达,分析miR-10b表达水平与患者临床特征之间的关系,为miR-10b后续功能研究奠定基础。方法提取35例手术切除的肾癌及癌旁正常组织标本中的总RNA,反转录获得cDNA,实时荧光定量PCR(qRT-PCR)检测miR-10b的表达水平,统计分析其表达水平与患者临床病理特征之间的关系。结果 35例组织标本中26例(74.29%)肾癌组织miR-10b表达水平下降(P<0.05),且下降和患者年龄、性别、肿瘤分期分级等无明显相关(P>0.05)。结论 miR-10b在肾癌组织中表达明显下调,提示其与肾癌发生有一定关系。 Objective To detect the expression levels of miR-10b in 35 RCC tissues and paired normal tissues and explore the relationship between the miR-10b expression and clinical characteristics of patients for further study of the function of miR-10b in RCC. Methods Total RNA isolated from 35 paired tissues obtained from nephrectomy were used to get eDNA by performing reverse transcription. The expression level of miR-10b was detected by quantitative real-time PCR (qRT- PCR) and the relationships with clinical characteristic were explored by statistical analysis. Results 26 cases (74.29%) showed down-regulation of miR-10b (P〈0.05). No relationships were found between the expression of miR-10b and patient's age, clinical stage or grade. Conclusions miR-10b is down-regulated in RCC, indicating it may be involved in the tumorigenesis of RCC.
