摘要
目的采用高效液相色谱-电喷雾质谱联用技术(LC-ESI-MS)对山竹果乙醇提取物中的化学成分进行分析鉴定,筛选山竹果中α-葡萄糖苷酶及黄嘌呤氧化酶抑制剂。方法选用反相C18色谱柱,以0.02%醋酸溶液(A)和乙腈(B)作为液相色谱流动相,进行梯度洗脱,流动相梯度程序:0~2 min,15%B,2~40 min,13%~50%B;流速0.6 ml/min;利用电喷雾质谱(ESI-MS)负离子模式在线检测化学成分;以α-葡萄糖苷酶及黄嘌呤氧化酶作为生物靶分子,以超滤质谱技术筛选酶抑制剂。结果共鉴定出5个化学成分,分别为3-异倒捻子素、Mangostanol、8-desoxygartanin、Gartanin和α-倒捻子素。该5个化合物均可与黄嘌呤氧化酶亲和,除Gartanin外均可以与α-葡萄糖苷酶亲和。结论利用液质联用技术对山竹果中化学成分进行分析可以快速分析鉴定山竹果中的化合物并提供结构信息。利用超滤质谱技术筛选有效成分具有快速和灵敏等优势。
Objective To analyze and identify the chemical compositions from Garcinia mangostana crude extracts by liquid chromatography coupled with electrospray ionization mass spectrometry( LC- ESI- MS),and screening the α- glucosidase and xanthine oxidase inhibitors from G. mangostana. Methods The analysis was processed by means of a reversed phase C18 column and a binary mobile phase system consisting of methanol and 0. 02% acetic acid aqueous under a gradient condition,Linear gradients:0- 2 min,13% B,2- 40 min,13%- 50% B; Electrospray ionization mass spectrometry( ESI- MS) under the negative ion mode to detect the chemical constituents. α- glucosidase and xanthine oxidase were employed as the target enzyme,and the ultrafiltration MS technique was employed to screen the inhibitors. Results Five chemical compounds from G. mangostana including 3- isomangostin,Mangostanol,8- desoxygartanin,Gartanin and α- mangostin respectively were separated and detected. The chemical compounds mentioned above all could be affinity with xanthine oxidase,and the chemical compounds mentioned above except of gartanin could be affinity with α- glucosidase. Conclusion The technique of HPLC / MS can quickly analyze and identify the compounds in Garcinia mangostana and provide structural information. Further more,screening the active compounds by using of ultrafiltration MS have the advantage of rapid and sensitive.
