摘要
目的对桑白皮蜜炙前后总黄酮提取物进行HPLC指纹图谱研究,探索总黄酮提取物在蜜炙前后质与量的变化。方法采用Waters 2965高效液相色谱仪进行检测,色谱柱Thermo ODS-2 HYPERSIL(250 mm×4.6 mm,5μm),色谱条件:乙腈-0.2%磷酸水溶液梯度洗脱;检测波长:PDA定时波长(0~30min,270nm)(30~90min,320nm);柱温30℃;流速1 ml·min-1;进样量10μl;分析时间:90 min。结果桑白皮与由其蜜炙而得蜜桑白皮总黄酮提取物指纹图谱相似度均大于0.975,而同一产地批次不相对应的桑白皮与蜜桑白皮总黄酮提取物指纹图谱相似度为0.528~0.856;中试样品蜜炙前后的相似度平均为0.988。结论桑白皮蜜炙前后差异较小,而相同产地市售桑白皮与蜜桑白皮之间存在较大差异,说明蜜炙工艺参数不统一对饮片内在质量的影响较大。
Objective Study on the HPLC fingerprint of total flavonoids extracted from Mori Cortex and Mori Cortex baked with honey,to explore changes in the quantity and quality of total flavonoids between Mori Cortex and Mori Cortex baked with honey.Methods Using Waters 2965 high performance liquid chromatography to detect and the chromatographic column was Thermo ODS- 2 HYPERSIL( 250 mm × 4. 6 mm,5 μ m). Chromatographic conditions acetonitrile- 0. 2% phosphate aqueous gradient elution,column temperature was 30 ℃,the flow rate was 1 ml·min- 1,the detection wavelength was PDA timing wavelength( 0 ~30min,270nm)( 30 ~ 90 min,320nm),the injection volume was 10 μl and the analysis time was 90 minutes. Results The total flavonoids fingerprint similarities were more than 0. 975 of Mori Cortex before and after baked with honey,and the similarities were0. 528 to 0. 856 of Mori Cortex and Mori Cortex baked with honey which not corresponded with each other. The average of the similarities was 0. 988 in the test samples before and after baked with honey. Conclusion The difference is small between Mori Cortex and Mori Cortex baked with honey,and there was large difference between Mori Cortex and Mori Cortex baked with honey sold in the same habitat area. The research shows that the honey process parameters were not unified,which had great influence on the intrinsic quality.
