摘要
建立金黄色葡萄球菌LAMP可视化检测方法。通过对2套引物的筛选,最终选取nuc基因的6条引物建立LAMP反应体系。整个反应在63℃恒温条件下进行45min,即可通过肉眼直接对反应结果进行判定。对金黄色葡萄球菌的检测灵敏度为0.001ng/μL,且与其他致病菌无交叉反应。该方法快速、特异性高,操作简便,适合基层快速检测。
To develop a loop-mediated isothermal amplification(LAMP)assay for visual detecting Staphylococcus aureus,2sets of specific primers were designed and 6primers targeting the nuc gene was selected.The method could detect Staphylococcus aureus within45 min at a constant temperature of 63 ℃.The sensitivity of the method was 1pg/μL,and had no cross reaction with other pathogens.The result indicated that the LAMP assay was a rapid,specific and simple tool for field diagnosis of Staphylococcus aureus.
