摘要
目的:通过骨组织Dlx5 mRNA及蛋白表达,探讨绝经后骨质疏松症(PMOP)的发病机制以及补肾壮骨中药复方的疗效机理。方法:去卵巢复制骨质疏松症大鼠模型按随机数字表法分为正常组、模型组、假手术组、补肾组、钙尔奇D组、骨疏康组,灌胃12周检测骨密度、Dlx5 mRNA及蛋白表达。结果:与正常组比较,模型组股骨骨密度明显降低,骨组织Dlx5 mRNA及蛋白表达明显降低;与模型组比较,补肾组、骨疏康组股骨骨密度明显升高,补肾组、骨疏康组骨组织Dlx5 mRNA及蛋白表达明显上调。结论:PMOP的发病机制之一可能是骨组织Dlx5 mRNA及蛋白表达降低,补肾壮骨中药复方可能通过上调骨组织Dlx5 mRNA及蛋白表达,有效防治绝经后骨质疏松症。
Objective: To discuss the pathogenesis of Postmenopausal Osteoporosis( PMOP) and the therapeutic effect and mechanism of reinforcing kidney to strengthen bone traditional Chinese medicine compound by mRNA and protein expression of Dlx5 in bone tissues. Methods: We established the osteoporosis rat models by ovariotomy. We divided all the rats into normal group,model group,sham-operated group,reinforcing kidney group,GAIERQI D group and GUSHUKANG group. After 12 weeks of gavage,we detected bone mineral density,mRNA and protein expression of Dlx5. Results: 1Compared with normal group,femur bone mineral density of model group decreased significantly,mRNA and protein expression of Dlx5 decreased significantly in bone tissues. 2 Compared with model group,femur bone mineral density of reinforcing kidney group and GUSHUKANG group increased significantly; mRNA and protein expression of Dlx5 of reinforcing kidney group and GUSHUKANG group up-regulated significantly in bone tissues. Conclusions: Maybe it is one aspect of the pathogenesis of PMOP that mRNA and protein expression of Dlx5 decreases in bone tissues; Reinforcing kidney to strengthen bone traditional Chinese medicine compound can prevent and treat PMOP by up-regulating mRNA and protein expression of Dlx5 in bone tissues.
