摘要
[目的]从陕西定边盐湖高渗极端环境中筛选分离获得嗜盐微生物,并对筛选获得的微生物进行种属鉴定.[方法]通过高盐选择性培养基平板分离法,从陕西定边盐湖土壤样品中,分离筛选获得极端嗜盐菌株,该菌最适生长温度35℃,最适pH值8.0.通过传统的表型特征(包括形态学观察、生长因子试验、生理生化特征)和系统发育学16S rDNA序列分析鉴定嗜盐菌株的分类学地位"结果]获得一株最适生长盐浓度在10%~20%的细菌菌株A426,初步鉴定其为盐单胞菌科(Halomonadaceae)盐单胞菌属(Hdomonas)菌株.[结论]成功获得定边盐湖地区中度嗜盐菌株A426.
[Objective] Halophilic bacteria strain was isolated from Dingbian salt lake sample in Shaanxi,and species indentification was studied. [Methods] By high salt selected culture method and gradient dilution isolation method isolate a halophilic bacteria strain from the Dingbian salt lake sample. The optimum growth temperature of it was 35 ~C ,the optimum pH value was 8.0.According to the classic phenotypic characteristics which include morphological observation,growth factor and the physiological and bio-chemieal characteristics,moderate halophile strain A426 was to be identified. To determine the taxonomy position of the newly isolated moderate halophile bacterium in evolution history, 16S rRNA sequence homologous alignment were performed between it and related species.[Resuhs] The strain A426 which could grow optimally in culture media that contain 10%-20% NaC1. The moderate halophile strain A426 was identified to belong to Halomonas sp.[Conclusion] Successfully isolate a moderate halonhile strain A426 from Dingbian salt lake.
