摘要 目的:基于ITS2序列鉴定十大功劳属药用植物,以避免该属物种药用基原混乱。方法:收集十大功劳属物种作为实验材料,采用改良的CTAB法提取总DNA,经PCR扩增、双向测序和序列拼接,应用MEGA 6.0对该属13个物种43条ITS2序列进行序列变异分析,计算种内种间Kimura 2-parameter(K2P)距离,构建系统邻接(NJ)树。结果:十大功劳属13个物种43条ITS2序列分析结果表明,各物种种内最大(平均)K2P距离均不大于种间最小(平均)K2P距离;系统NJ树结果显示,十大功劳属其中9个物种各自聚为一支。结论:ITS2序列对十大功劳属药用植物具有较好鉴定效果,为该属药用植物的临床用药安全提供了分子依据。 Objective:To Identify the genus Mahonia medicinal plants,based on ITS2 sequences.Methods:Total genomic DNA was extracted from experimental materials by the improved CTAB method.The ITS2 sequences were obtained after PCR amplification,bidirectional sequencing and sequence assembly.The genetic distances based on the Kimura 2-parameter(K2P)model,variable sites and neighbor-joining(NJ)phylogenetic tree were computed by MEGA 6.0.Results:Based on ITS2 sequences,the study indicated that the intraspecific maximum and average genetic distances were no greater than the interspecific ones.In all of the 13 samples,9 of them were respectively clustered into a single branch in the NJ phylogenetic tree.Conclusion:ITS2 sequence has a good effect on the identification of the genus Mahonia medicinal plants,which can provide basis to ensure the safety of medicines of the genus Mahonia medicinal plants.
