摘要
目的:通过ISSR分子标记技术考察北京地区野生柴胡不同种质资源之间的亲缘关系。方法:从43条引物中筛选出8条合适的引物,对北京地区不同产地采集的15份野生柴胡种质资源进行ISSR分析,构建聚类系统树状图。结果:8条引物共扩增出130条条带,其中多态性条带114条,占87.7%,聚类分析显示北京地区野生柴胡遗传多样性较高,并存在明显的种内遗传变异。结论:北京地区野生柴胡呈现一定的地域性分布趋势,应加以保护,初步认为百花山山腰及山脚柴胡值得推广种植,研究为北京地区柴胡种质亲缘关系研究及栽培品种的选育奠定了基础。
Objective:To assess population genetic diversity of different kinds of individuals of Bupleurum chinense in Beijing area by inter simple sequence repeat (ISSR) technique.Methods:Eight appropriate ones were selected from a total of forty-three primers for ISSR PCR amplification to analyse the Bupleurum in Beijing area and draw dendrograms by using genetic distance UPGMA method.Results:A total of 130 bands and 114 polymorphic bands were amplified by 8 primers,the polymorphic loci accounted for 87.7%.The cluster analysis showed that genetic diversity was high in Bupleurum, the intraspecific genetic variation was obvious.Conclusion:The distribution of Bupleurum germplasm in Beijing exhibited a certain regional characteristics.It is imperative to strengthen the protection of Bupleurum wild resources.The plants in the foot and hillside of Baihuashan is good for cultivating.This study laid a foundation for the research of relationship between Bupleurum and the breeding of cultivars.
