摘要
目的:进一步评价芪精多糖片治疗骨髓抑制小鼠的疗效,探讨其作用机制。方法:采用小鼠腹腔注射环磷酰胺造成化疗骨髓抑制模型,用芪精多糖片干预治疗,常规计数白细胞、红细胞、血小板、骨髓有核细胞数,流式细胞仪检测Sca-1+CD34+细胞以比较骨髓造血干细胞比例,FQ-PCR法检测Notch1、Notch2 mRNA表达。结果:治疗组白细胞、骨髓有核细胞、造血干细胞数、Notch1表达、Notch2表达均高于模型对照组,血小板、红细胞治疗组与模型组未发现差异。结论:芪精多糖片具有良好的升高化疗骨髓抑制小鼠白细胞的作用,其作用机理与调控Notch1、Notch2 mRNA表达、促进骨髓造血干细胞增殖有关。
Objective:To further evaluate the treatment efficacy of Qi Jing polysaccharide tablets on mice with bone marrow suppression and explore the mechanism.Methods:The mice bone marrow suppression model caused by intraperitoneal injection of cyclophosphamide were used in the study,Qi Jing polysaccharide tablets were given for the intervention treatment.The conventional counting white blood cells,red blood cells,platelet count,bone marrow nucleated cells were measured,the Sca-1+CD34+ positive cells of bone marrow were detected by using flow cytometry to compare the stem cells number.The Notch1 and Notch2 mRNA expression were detected by FQ-PCR method.Results:The Notch1,Notch2 mRNA expression in white blood cells,bone marrow cells and hematopoietic stem cell number in the bone marrow in the treatment group were higher than those in the control group.There was no difference between treatment group and the untreated group in platelets and red blood cells counting.Conclusion:Qi Jing polysaccharide tablets have good effect on increasing white blood cells of mice with chemotherapy caused myelosuppression,the mechanism is related to regulation of Notch1,Notch2 mRNA expression and promotion of bone marrow stem cell proliferation.
