摘要
目的研究microRNA-215(miR-215)在结直肠肿瘤组织和病人血清中的表达水平及其临床意义。方法从结直肠癌、结直肠腺瘤病人病变组织及健康查体者(正常对照组)结直肠组织和血清标本中提取微小RNA(microRNA),应用实时荧光定量PCR(RT-qPCR)方法检测miR-215的表达,分析miR-215表达水平与结直肠肿瘤临床病理参数间的关系,并通过绘制受试者工作曲线(ROC曲线)探讨血清miR-215在结直肠肿瘤诊断中的价值。结果与正常对照组比较,miR-215在结直肠癌和结直肠腺瘤病变组织中表达显著降低,差异有显著性(F=16.603,t=5.756、3.572,P〈0.01);miR-215在结直肠癌和结直肠腺瘤病人血清中的表达也低于正常对照组,差异有显著性(F=27.329,t=7.377、4.691,P〈0.01);与结直肠腺瘤组相比,结直肠癌组miR-215表达下调(t=2.182、2.671,P〈0.05)。结直肠癌组织和病人血清中miR-215的低表达与病人的临床分期有关(F=4.073、6.125,P〈0.05),且结直肠癌组织与血清中miR-215的表达量呈正相关(r=0.696,P〈0.01);miR-215表达与结直肠腺瘤临床特征无关(P〉0.05)。ROC曲线分析显示,结直肠癌和正常对照组血清miR-215的ROC曲线下面积(AUC)为0.855(95%CI=0.740~0.970),结直肠腺瘤与正常对照组血清miR-215的AUC为0.774(95%CI=0.634~0.913)。结论 miR-215在结直肠癌的早期诊断中具有潜在的价值。
Objective To explore miR-215 expression in tumor tissues and in serum in patients with colorectal neoplasms and its clinical significance. Methods MicroRNA was extracted from tissues of colorectal cancer,colorectal adenoma and healthy individuals who came for medical examination.The expression of miR-215 was detected using quantitative real-time polymerase chain reaction(RT-qPCR).The relationship between the level of miR-215 expression and clinicopathological parameters in colorectal neoplasms was analyzed,and the value of serum miR-215 in the diagnosis of colorectal tumors was evaluated through drawing a receiver operating characteristic(ROC)curve. Results Compared with normal controls,the expression of miR-215 in tissues of colorectal cancer and colorectal adenoma was lower(F=16.603;t=5.756,3.572;P〈0.01),and that in serum was also lower(F=27.329;t=7.377,4.691;P〈0.01).Compared with colorectal adenoma,the expression of miR-215 in colorectal cancer was downregulated(t=2.182,2.671;P〈0.05).Low expression of miR-215 in cancer tissue and serum was associated with clinical stage of the disease(F=4.073,6.125;P〈0.05),and the expression of miR-215 in cancer was positively correlated with that in serum(r=0.696,P〈0.01),but the expression of miR-215 was not correlated with clinical features of colorectal adenoma(P〉0.05).ROC curve analysis demonstrated that the area under the ROC curve of serum miR-215 in the cancer and normal control group was 0.855(95%CI=0.740-0.970)and that in the adenoma and normal control was 0.774(95%CI=0.634-0.913). Conclusion There is a potential value of miR-215 in early diagnosis of colorectal cancer.
