联合使用免疫调节剂后对复发性生殖器疱疹豚鼠模型免疫状态的影响 被引量:3

The Influence of Combined Application of Immunomodulator in Different Periods of Time to the Immune State of RGH Guinea Pig Model

摘要 目的探讨不同时间段联合使用铜绿假单胞菌注射液及匹多莫德片治疗后复发性生殖器疱疹(RGH)豚鼠模型的免疫状况,借以间接揭示人类RGH患者长期间隙性服用免疫调节剂的疗效。方法用HSV-2 Sav感染已排除HSV感染的雌性中华彩豚,造成RGH豚鼠模型后,用双抗体夹心酶联免疫法(ELISA)分别先后测出不同用药时间段(3个月,6个月,12个月)、RGH豚鼠模型组(未给药)及空白对照组的豚鼠血清IL-5,10,13及血清IFN-γ的浓度(pg/m L),再分别进行比较。结果 RGH豚鼠模型组IL-5,10及IL-13的浓度均显著高于空白对照组,不同用药时间段豚鼠的IL-5,10及IL-13浓度均低于RGH豚鼠模型组,差异有统计学意义(P<0.01),而RGH豚鼠模型组IFN-γ的浓度显著低于空白对照组,不同用药时间段豚鼠的IFN-γ浓度均高于RGH豚鼠模型组,相比较差异有统计学意义(P<0.05)。结论不同时间段联合使用铜绿假单胞菌注射液及匹多莫德片后可纠正RGH豚鼠模型机体细胞免疫与体液免疫的分化状态。 Objective To explore the immune state of recurrent genital herpes ( RGH ) guinea pig model after combined application of Pseudomonas Aeruginosa injection and Pidotimod in different periods of time to, and reveal the therapeutical effect of immunomodulator when applying to human RGH patients long-term intermittently. Methods We gained the RGH guinea pig model by using HSV-2 sav to infect female guinea pig which had never been infected by HSV infection. The double antibody sandwich enzyme-linked immunosorbent assay(ELISA) was used to test the concentration (pg/mL) of IL-5, IL-10, IL-13 and IFN-gamma in guinea pig serum which got from the different medication period group ( sampled from 3 months, 6 months and 12 months ), RGH guinea pig model group(nonedication ) and blank control group. We compared the data between each group. Results Serum IL-5,10 and IL-13 levels of RGH guinea pigs model group were significantly higher than blank control group. Serum IL-5,10 and IL-13 levels of guinea pigs in different drug use time were all lower than RGH guinea pig model group, with significant differences( P 〈 0. 01 ). While IFN- gamma concentration of RGH guinea pigs model were significantly lower than blank control group, and IFN- gamma concentration of guinea pigs in different drug use time were all higher than RGH guinea pig model group, with significant differences ( P 〈 0. 05 ). Conclusion Combined application of Pseudomonas Aeruginosa injection and Pidotimod in different periods of time can correct differentiation state of cellular and humoral immunity of RGH guinea pig model.
