摘要
目的探讨糖肾宁对自发性2型糖尿病KK-Ay小鼠肾组织肝细胞生长因子(hepatocyte growth factor,HGF)表达的影响。方法采用自发性2型糖尿病KK-Ay小鼠建立糖尿病肾病模型,根据小鼠随机血糖,采用分层随机法分为模型组、缬沙坦组和糖肾宁组,以C57BL/6J小鼠作为空白组,每组均为20只。药物连续干预12周,测定糖化血红蛋白(hemoglobin A1c,Hb A1c)、血清尿素氮(blood urea nitrogen,BUN)、血清肌酐(serum creatinine,SCr),采用Western blotting法检测肾组织HGF蛋白表达水平。结果与空白组比较,模型组Hb A1c、BUN、SCr明显升高(P<0.05),肾组织HGF表达明显降低;与模型组比较,缬沙坦和糖肾宁组Hb A1c无明显变化(P>0.05),肾组织HGF表达明显升高,BUN、SCr均有不同程度的降低(P<0.05),缬沙坦和糖肾宁组比较,差异无统计学意义(P>0.05)。结论糖肾宁可降低糖尿病小鼠BUN、SCr,升高肾组织HGF蛋白表达量,抑制肾脏纤维化,具有防治糖尿病肾病的作用。
Objective To study the effects of Tangshenning on the protein expression of Hepatocyte Growth Factor( HGF) in KK-Ay mice with spontaneous type 2 diabetic mellitus( T2DM). Methods Diabetic male KK-Ay mice were randomly divided into three groups:the model group,the valsartan group and the Tangshenning group,with 20 in each. Twenty male C57 BL /6J mice were chosen as the normal group. After intervention with Tangshenning and valsartan for 12 weeks,hemoglobin A1c( Hb A1c),blood urea nitrogen( BUN)and serum creatinine( SCr) were examined. Expression of HGF was assessed by Western blotting. Results Compared with blank group,Hb A1 c,BUN and SCr in model group are significantly increased( P〈0. 05) while the expression of HGF was significantly decreased.Compared with model group,there were no significant changes in the levels of Hb A1c( P〉0. 05). The mean levels of BUN and SCr in both treatment group were lower than model group( P〈0. 05). The expression of HGF was significantly increased. Conclusion Tangshenning can decrease BUN,SCr and prevent DN by promoting the expression of HGF.
