优化中药复方“松友饮”的基础研究

摘要 目的:研究中药复方"松友饮"的体内药效物质基础并重新配伍设计,为其组方优化提供理论基础。方法:应用高转移人肝细胞癌细胞株MHCC97H构建裸鼠原位移植瘤模型,24只成模裸鼠随机平均分成对照组和"松友饮"组,肝脏接种肿瘤1 d开始1次/d灌胃给药("松友饮"3.6 g/kg,对照组予以等量蒸馏水)。灌胃5周,脱颈处死裸鼠收集血样,用于进一步分析"松友饮"体内药效物质基础,采用均匀设计的实验方法重新设计、配伍、优化"松友饮"。结果:裸鼠模型构建过程顺利,灌胃过程中实验组死亡1只裸鼠,有效获取含药血清。结合参考文献及通过比对发现连续给药5周,"松友饮"吸收入血的主峰来自组方药材成分:黄芪甲苷、槲皮素及隐丹参酮、丹参酮IIA(新组复方主要含4种单体,简称HHYD复方),重新配伍为3种新组方。结论:基于5周给药的含药血清药效物质基础新组方的组分更加明确,更易定量和质量控制。 Objective: To investigate the in vivo effective substances of Chinese herbal compound Songyou Yin and to provide the experimental basis for its optimization. Methods: Metastatic human hepatocellular carcinoma( HCC) nude mice models were established by orthotopic implantation with human HCC cell line MHCC97 H. Twenty- four nude mice bearing orthotopic xenografts were randomized into control group and Songyou Yin group. Daily intragastric administration of Songyou Yin( 3. 6 g / kg) was given first 24 hours after orthotopic implantation. Mice were sacrificed by cervical dislocation to collect serum sample at 5th week for further analyses of effective substances of Songyou Yin. The uniform design experimental method was applied for redesign,compatibility and optimization of Songyou Yin. Results: Metastatic human HCC nude mice models were successfully established and medicated serum was collected. During the therapy,there was only one mouse died. The predominant peaks of Songyou Yin derived from those components include Astragaloside IV,Quercetin,Cryptotanshinone and Tanshinone IIA( New formula,HHYD). Conclusion: Based on 5 weeks therapeutic substances of medicated serum of Songyou Yin,we got a new formula that is more precise,more quantitative and understand its quality control.
