半夏泻心汤拆方对胃溃疡大鼠胃黏膜修复因子EGF、VEGF表达的影响及方中寒热并用配伍意义的研究

Effect of Decomposed Banxia Xiexin Decoction on Repair Factors EGF and VEGF in Gastric Mucosa of Rats with Gastric Ulcer Model and Its Significance of Compatibility of Both Cold And Heat

目的:观察半夏泻心汤拆方对胃溃疡大鼠模型胃黏膜修复因子EGF、VEGF表达的影响,分析该方治疗胃溃疡的作用机制,探讨该方中寒凉、温热、补益不同属性药物在治疗胃溃疡中的配伍意义,揭示本方中寒热并用、补泻同施的配伍方法的优势和作用靶点。方法:SD大鼠,雌雄各半,造模各组采用大承气汤结合辣椒/乙醇混合液灌胃及乙酸注射法建立胃溃疡寒热错杂证病证结合动物模型。胃溃疡手术24h后给予半夏泻心汤及拆方各组药物灌胃,28d后处死大鼠,采集胃组织,免疫组化染色法检测胃黏膜修复因子EGF、VEGF的表达。结果:与空白组比较,模型组大鼠胃黏膜EGF、VEGF平均光密度值明显升高,差异有统计学意义(P<0.01);与模型组比较,全方组、寒热并用组、补益组、温补组、寒补组及阳性组EGF、VEGF平均光密度值显著升高(P<0.01),温热组升高(P<0.05),而寒凉组无显著性差异(P>0.05);与全方组比较,寒热并用组、补益组、温补组、寒补组及阳性组大鼠胃黏膜EGF、VEGF平均光密度值无差异(P>0.05),而寒凉组和温热组有显著性差异(P<0.01)。结论:半夏泻心汤方中不同属性药物主要通过提高胃黏膜修复因子EGF、VEGF的表达从而对胃溃疡发挥治疗作用。寒热并用配伍能消除单用寒凉或温热药的局限和不足,在治疗方面产生了协同增效的作用。

Objective： To observe the effects of decomposed Banxia Xiexin decoction on the expressions of repair factors EGF and VEGF in gastric mucosa of rats with gastric ulcer model and analyze the mechanism of Banxia Xiexin decoction and its subdivisions,revealing the superiority and targets of compatibility of both cold and heat. Methods： Male and female SD rats were selected to establish the gastric ulcer animal model of mixed cold and heat syndromes. After 24 hours,all the rats except blank group and sham operation group were given Banxia Xiexin decoction and different herbal components. All rats were sacrificed after 28 days,observing rats＇ pathological morphology change of gastric tissue and testing gastric mucosal EGF and VEGF expressions by immunohistochemical detection. Result： Compared with blank group,the average optical density of EGF and VEGF expressions was significantly enhanced of model group rats（ P〈 0. 01）. Compared with model group,the average optical density of EGF and VEGF expressions were significantly enhanced of Banxia Xiexin decoction group,compatibility of both cold and heat group,tonic group,heat tonic group,cold tonic group and positive group（ P 〈0. 01）,while it was enhanced in heat group（ P 〈0. 05） and cold group had no significant difference（ P 〉0. 05）. Compared with Banxia Xiexin decoction group,compatibility of both cold and heat group,tonic group,heat tonic group and cold tonic group had no significant difference（ P 〉0. 05） and cold group and heat group had significant difference（ P 〈0. 01）. Conclusion： The different properties drugs of Banxia Xiexin decoction play a therapeutic action on gastric ulcer by increasing the expression of gastric mucosa repair factors EGF and VEGF. Compatibility of cold and heat drugs can eliminate their adverse effects and enhance the cooperative effect.
