中药白秓合剂对HaCaT细胞CXCR2蛋白表达及TNF-α诱导的HaCaT细胞IL-6表达的影响 被引量:1

摘要 目的:观察中药白疕合剂对人永生化角质形成细胞(Ha Ca T细胞)CXC趋化因子受体2(CXCR2)蛋白表达及肿瘤坏死因子(TNF)-α诱导的Ha Ca T细胞分泌白细胞介素(IL)-6和IL-6 mRNA表达的影响,并探讨其作用机制。方法:用白疕合剂干预Ha Ca T细胞,Western-blot检测CXCR2蛋白表达;用白疕合剂干预TNF-α诱导的Ha Ca T细胞,ELISA检测IL-6分泌量,RT-PCR检测IL-6 mRNA表达。结果:白疕合剂中、高剂量组对CXCR2蛋白的表达有明显的抑制作用;白疕合剂低、中、高剂量组对IL-6分泌量和IL-6 mRNA表达均有明显的抑制作用且呈浓度依赖性。结论:白疕合剂可能通过抑制IL-6分泌,降低IL-6 mRNA、CXCR2蛋白表达发挥对银屑病的治疗作用。 Objective: To observe the effects of white mange mixture on the expression of CXC chemokine receptor 2( CXCR2) protein of Ha Ca T cells and the content of IL- 6 and the expression of IL- 6 mRNA of Ha Ca T cells induced by TNF- α and to explore its mechanism. Methods: Using white mange mixture intervening Ha Ca T cells,Western blot was used to detect the expression of CXCR2 protein and using white mange mixture interventing Ha Ca T cells induced by TNF- α. ELISA was applied to test the the content of IL- 6 and the expression of IL- 6mRNA was detected by PT- PCR.Results: White mange mixture( middle and high dose groups) could obviously inhibit the expression of CXCR2 protein and low,middle and high dose groups could obviously inhibit the content of IL- 6 and decrease the expression of IL- 6 mRNA in a dose- dependent manner. Conclusion: White mange mixture may inhibit the content of IL- 6 and decrease the expression of IL- 6 mRNA and CXCR2 protein to play a role in the treatment of psoriasis mechanism.
