摘要
目的:吴茱萸次碱(Rutaecarpa RUT)是芸香科植物吴茱萸的主要成分,它可激活辣椒素受体促进降钙素基因相关肽(CGRP)等神经递质的释放来发挥药理作用。本实验的目的为观察吴茱萸次碱软膏对小鼠银屑病模型治疗作用。方法:通过药剂学方法制成了不同浓度(2%,5%,10%)RUT软膏,在小鼠尾部与背部银屑病模型给药14天,观察小鼠尾部颗粒层形成的变化,给药结束后取小鼠背部皮肤组织匀浆后采用放射免疫方法检测CGRP水平变化,并且取小鼠血浆检测CGRP水平变化。结果:不同浓度的吴茱萸次碱软膏均能促进小鼠尾部颗粒层形成(P<0.05),并且浓度为5%与10%的RUT软膏能显著降低小鼠背部CGRP含量(P<0.05),但是其对小鼠血浆的CGRP没有影响。结论:吴茱萸次碱外用对小鼠银屑病模型有一定的治疗作用,这种作用与其促进小鼠皮肤CGRP释放而导致其含量降低有关。
Objective: Rutaecarpa(RUT) is the main component Rutaceae Evodia, which can activate the capsaicin receptor to promote the release of calcitonin gene-related peptide(CGRP) and/or other neurotransmitters, in turn exert pharmacological effects. The purpose of this experiment is to observe rutaecarpine ointment treatment effect on psoriasis mouse model and approach its mechanism.Methods: Different concentrations(2%, 5%, 10%) RUT ointments were prepared by pharmaceutical method. Mice tail particle layer forming were chosen as psoriasis model, and mice skin of tail and back were administered by RUT ointments for 14 days to observe the changes of mouse tail granular layer forming or skin CGRP levels. At the end of the administration mice back skin tissue homogenates were determined CGRP levels using radioimmunoassay, and plasma of mice were taken to detect in CGRP levels. Results: Different concentrations of rutaecarpine ointment promote mouse tail granular layer(P〈0.05), and a concentration of 5% and 10% of the RUT ointment can significantly reduce the back skin of mice CGRP content(P〈0.05). Conclusion: Rutaecarpine exert therapeutic effect on mouse psoriasis model, which is related to exhausting mouse skin CGRP.
