Orexin-A受体介导的生长抑素激动剂ODT8-SST对大鼠摄食和饮水的影响

Effect of Orexin-A Receptor Mediated Somatostatin Agonists ODT8-SST of Rats on Food Intake and Water Intake

摘要 目的:探讨orexin-A(OXA)受体介导的生长抑素激动剂ODT8-SST对大鼠摄食和饮水的调节作用相关作用机制。方法:在光照周期内,大鼠40只随机分8组,侧脑室(icv)分别注射不同剂量ODT8-SST或生理盐水(NS);大鼠56只随机分8组分别侧脑室注射不同剂量OXA受体(OX1R)拮抗剂SB-334867或NS;2小时后测量大鼠摄食量和饮水量。结果:与NS组相比,实验组大鼠侧脑室注射ODT8-SST(1μg/rat),2小时后摄食量和饮水量均显著增加(P<0.05)。大鼠侧脑室注射SB-334867(16μg/rat)完全抑制了由侧脑室注射ODT8-SST后引起的摄食量和饮水量的增加;与此相反,大鼠给予SST2拮抗剂S-406-028预处理之后,可阻止侧脑室注射ODT8-SST引发的促进食欲作用,但不会影响侧脑室注射OXA(10.7μg/rat)诱导的摄食量和饮水量的增加。结论:侧脑室注射ODT8-SST可促进摄食和饮水,该过程可能由OX1R所介导;orexin-A促进摄食作用不依赖大脑SST2通路的激活。 Objective: To investigate the orexin-A(OXA) receptor mediated somatostatin agonists ODT8-SST on the regulation of food intake and water intake of rats and OXA promoting feeding mechanism. Methods: In the light period, 40 rats were randomly divided into 8 groups, the lateral ventricle(icv) were injected with different doses of ODT8-SST or normal saline(NS); 56 rats were randomly divided into 8 groups, icv were injected with different doses of OXA receptor(OX1R) antagonist SB-334867 or NS; the food intake and water intake of rat were recorded for 2 hours after the injection. Results: Compared with NS control group, after injecting ODT8-SST(1μg / rat, icv) into ICV, the food intake and water intake of experimental group were significantly increased(P 0.05). Lateral ventricle injection of OXA receptor(OX1R) antagonist SB-334867(16 μg / rat) completely inhibited the increase in food intake and water intake caused by lateral ventricle injection of ODT8-SST; with in contrast, rats were given SST2 antagonist S-406-028 after pretreatment, can prevent the promotion of the role of appetite caused by intracerebroventricular injection of ODT8-SST, but does not affect the lateral ventricle injection OXA(10.7 μg/rat) induced food intake and water intake increased. Conclusions: Intracerebroventricular injection of ODT8-SST can promote feeding and drinking, OX1 R signaling pathway is a part of the neural circuits; orexin-A promotes the ingestion does not depend on the activation of the brain SST2 pathway.
