根皮素对HepG2肝癌细胞增殖和凋亡影响的实验研究 被引量:1

摘要 目的:探讨根皮素对人肝癌Hep G2细胞增殖和凋亡的影响。方法:MTT法检测不同浓度(30、40、50μg/m L)根皮素对肝癌Hep G2细胞增殖的抑制作用,流式细胞技术检测根皮素对Hep G2细胞凋亡的影响,ELISA法检测不同浓度根皮素干预后细胞中Bcl-2和Bax表达的变化,Western blot法检测30μg/m L根皮素在8,16,24小时后检测p-AKT蛋白表达情况。结果:30、40和50μg/m L的根皮素对肝癌Hep G2细胞的增殖均有抑制作用(P<0.01),同时,30、40和50μg/m L的根皮素在24小时后可诱导Hep G2细胞发生早期凋亡,凋亡率分别为0.1321±0.0224,0.2607±0.0457,0.3712±0.0884(P<0.01);另外,30、40和50μg/m L的根皮素作用24小时后细胞内Bcl-2表达降低,Bax表达增高(P<0.01);最后,30μg/m L根皮素可以明显减少p-AKT表达量,这种作用呈现时间依赖性。结论:根皮素能够抑制肝癌Hep G2细胞的增殖和促进细胞凋亡。 Objective: To evaluate the effects of phloretin on proliferation and apoptosis of Hep G2 cells. Methods: Effects of phloretin on Hep G2 cell proliferation and apoptosis were evaluated by MTT method and flow cytometry instrument, respectively.Expression of Bcl-2 and Bax protein in Hep G2 cell with or without phloretin pretreatment was detected by ELISA method. Expression of the p-AKT protein in Hep G2 cell which were treated with phloretin(30 μg/m L) in 8, 16, 24 hours can be measured by Western Blot.Results: Different doses of phloretin(30, 40 and 50 μg/m L) depressed the proliferation of Hep G2 cells(P〈0.01). Besides, all dosages of phloretin(30, 40 and 50 μg/m L) induced cell apoptosis in Hep G2 significantly compared to that of control,the relative rate of apoptosis:0.1321±0.0224, 0.2607±0.0457, 0.3712±0.0884(P〈0.01). What was more, 30, 40 and 50 μg/m L phloretin decrease the expression of Bcl-2 and increased Bax expression, respectively; and phloretin can decrease the expression of the p-AKT. Conclusion: Phloretin could inhibit proliferation and induced apoptosis on Hep G2 cells.
