摘要
目的研究急性复合应激模型大鼠心肌组织中B型钠尿肽(BNP)的表达,探讨其与急性应激导致心脏性猝死的关系。方法将25只雄性SD大鼠随机分为2组,模型组15只,对照组10只。模型组大鼠采用束缚加电击的急性复合应激方法造模。应用化学发光法检测两组大鼠血清皮质醇的含量,免疫组化方法检测大鼠心肌组织中BNP蛋白的表达。结果模型组大鼠血清皮质醇含量明显高于对照组(t=8.78,P<0.05),显示模型组大鼠造模成功。模型组大鼠心肌细胞胞浆呈免疫组化阳性着色,其心肌组织BNP蛋白表达(0.169±0.279)高于对照组(0.144±0.572),差异有统计学意义(t=2.55,P<0.05)。结论 BNP可能参与了急性复合应激导致心脏性猝死的过程,检测该因子的变化可为应激导致心脏性猝死的法医学鉴定提供新的思路。
Objective To investigate the expression of brain natriuretic peptide(BNP)in rat myocardial tissue of a rat model of acute compound response and explore the relationship between BNP and sudden cardiac death due to acute stress. Methods Twenty-five male SDrats were randomized to two groups as model group(n=15)and control group(n=10).A rat model was created using combined stress of restraint and electric shock.Using chemiluminescence to detect the serum cortisol content,and immunohistochemistry to detect the expression of BNP protein in myocardial tissue. Results The serum cortisol content of rats in the model group was significantly higher than that in the control group(t=8.78,P〈0.05),indicating success of the modelling.Immunohistochemistry showed positive staining of myocardial cytoplasm and the expression of BNP protein was(0.169±0.279),higher than that in the control group(0.144±0.572)(t=2.55,P〈0.05). Conclusion Brain natriuretic peptide probably involves in the process of sudden cardiac death caused by acute stress.Detecting the changes of the factor can provide a new idea in medicolegal identification for this condition.
