摘要
目的探讨顺铂联合姜黄素灌胃对裸鼠皮下移植瘤生长及瘤组织增殖细胞核抗原(PCNA)和基质金属蛋白酶2(MMP2)表达影响。方法采用PC3细胞株制成裸鼠皮下移植瘤模型,随机分为溶媒对照组、姜黄素组、顺铂组、顺铂+姜黄素组。治疗20d后,比较各组小鼠肿瘤体积大小、去瘤体质量,并采用荧光定量RT-PCR方法检测瘤组织中PCNA和MMP2 mRNA的表达。结果顺铂+姜黄素组小鼠体质量明显大于顺铂组(F=19.12,q=6.02,P〈0.01)。治疗第20天时,顺铂+姜黄素组肿瘤体积明显小于顺铂组和姜黄素组(F=52.93,q=4.87、16.25,P〈0.01)。顺铂+姜黄素组肿瘤组织中PCNA及MMP2mRNA表达量均明显少于顺铂组和姜黄素组(F=132.45、47.44,q=6.36~27.88,P〈0.01)。结论顺铂联合姜黄素灌胃可共同抑制裸鼠皮下移植瘤生长增殖。
Objective To investigate the impact of intragastric administration of combined cisplatin with curcumin on the growth of transplanted tumors in nude mice and the expressions of proliferating cell nuclear antigen(PCNA)and matrix metalloproteinase 2(MMP2)in tumor tissue. Methods Models of PC3 transplanted tumor were created in nude mice and randomized to solvent-control group,curcumin group,cisplatin group and cisplatin+curcumin group.After 20 days of treatment,the tumor volume and net body weight were compared between each group and the expressions of PCNA mRNA and MMP2 mRNA were detected using RT-PCR. Results The body weight of mice in cisplatin+curcumin group was heavier than that in cisplatin group(F=19.12,q=6.02,P〈0.01).On day 20 after the treatment,the tumor volume in the cisplatin+curcumin group was smaller than that in cisplatin and curcumin groups(F=52.93;q=4.87,16.25;P〈0.01),and the expressions of PCNA mRNA and MMP2 mRNA decreased(F=132.45,q=10.14-27.88,P〈0.01). Conclusion Intragastric administration of cisplatin plus curcumin can jointly inhibit the growth of subcutaneously transplanted tumors.
