中药单体黄芩苷对乙醇所致淋巴细胞DNA损伤的保护作用研究 被引量:2

摘要 目的:探讨中药单体黄芩苷对乙醇所致淋巴细胞DNA损伤的保护作用。方法:提取10名健康志愿者外周血中的淋巴细胞,与乙醇、黄芩苷孵育,考察中药单体黄芩苷对乙醇所致淋巴细胞DNA损伤的保护作用。结果:30 Mm乙醇作用淋巴细胞1 h导致尾距由0.20升高至0.78;黄芩苷单体不能抑制淋巴细胞增殖(P>0.05);黄芩苷组(20 ng·L-1)尾距阵显著低于空白组(纯乙醇)(P<0.05),且对乙醇所致淋巴细胞DNA损伤的保护作用呈浓度依赖性。结论:黄芩苷对乙醇所致的人体外周淋巴细胞DNA损伤具有一定的保护作用,这为黄芩苷等天然产物在抗DNA氧化损伤,治疗自由基等疾病方面提供药理学依据,具有进一步研究价值。 Objective :To explore monomeric astragaloside in protecting lymphocytes DNA damage due to alcohol. Methods:Ten healthy volunteers were chosen and their lymphocytes in the peripheral blood were extracted, and incubated together with alcohol and astragaloside to explore monomeric astragaloside in protecting lymphocytes DNA damage due to alcohol. Resuhs:After 30Mm had affected lymphocytes for one hour, microspur rose from 0.2 to 0.78. Monomeric astragaloside could not inhibit lymphocyte proliferation (P 〉 0.05 ). The microarray of the astragaloside group (20 μg ·mL-1) was much lower than that of the control group ( purealcohol) ( P 〈 0.05 ) , and deeply depend on the protective function for DNA damage caused by lymphocytes. Conclusion : Astragaloside has certain protective function for the peripheral blood lymphocytes DNA damage due to alcohol, which provides pharmacology evidences for astragaloside in resisting DNA oxidative damage and treatment of free radicals diseases, and is worth researching.
