摘要 目的:通过组织块分离法将冬凌草不同器官中内生真菌进行分离纯化,确定最优分离培养基及冬凌草中的内生真菌类型。方法:以冬凌草不同器官(叶片、茎)为实验材料,采用组织块分离法对冬凌草进行内生真菌的分离并纯化至单一菌落,保存备用。结果:现已从冬凌草叶片及茎中分离出内生真菌共256株,其中茎中分离出113株,叶片中分离出143株,其中PDA培养基共分离出内生真菌86株。结论:通过对冬凌草不同器官中内生真菌的分离及纯化,得到了冬凌草中内生真菌的类型,充分说明了冬凌草中内生真菌具有一定程度的生物多样性;同时确认PDA培养基为冬凌草内生真菌分离的最优培养基。 Objective:To separate and purify endophytic fungi from different organs of Rabdosia rubescer( Hemsl. )Hara. by using tissue separation method to determine the optimal separation euhure medium and types of endophytic fungi. Methods : Different organs( leaf, stem) in Rabdosia rubescens (Hemsl.)Hara. were used for experiment materials to separate and purity' its endophytic fungi to a single colony by using tissue separation method. Then they were set aside for future use. Results:The total 256 endo- phytic fungi were separated from its leaves and sterns(including 113 from stems and 143 from leaves) ,and 68 were separated by PDA culture medium. Conclusion : In this way, types of endophytic fungi in Rabdosia rubesce1 ( Hemsl. ) Hara. are determined, which shows variety of creature ; At the same time, PDA culture medium is regm, ded as the best one to separate its endophytic fungi.
