黄芪多糖对雄性肥胖小鼠精子蛋白酪氨酸磷酸酶1B表达及顶体反应的影响 被引量:3

Influence of astragalus polysaccharides on the expressions of protein tyrosine phosphatase 1B and acrosome reaction in sperm from male obese mice

摘要 目的探讨黄芪多糖对雄性肥胖小鼠精子蛋白酪氨酸磷酸酶1B(protein tyrosine phosphatase 1B,PTP1B)表达和顶体反应的影响。方法 40只C57BL/6J雄性小鼠随机分为对照组10只和高脂饲料组30只,分别给予普通饲料和高脂饲料喂养。8周后选取造模成功的肥胖小鼠20只,随机分为模型组和黄芪多糖(astragalus polysaccharides,APS)干预组(APS干预组),每组10只。对照组、模型组给予生理盐水灌胃,APS干预组给予APS 200mg/(kg·d)灌胃。4周后3组小鼠采用Western blot法检测精子PTP1B表达水平,采用金霉素染色法检测精子顶体反应率。结果高脂饲料组小鼠体质量[(32.8±1.7)g]明显高于对照组[(25.6±0.6)g](P<0.01);模型组小鼠精子顶体反应率[(10.6±6.2)%]明显低于对照组[(44.7±10.5)%]和APS干预组[(21.3±8.5)%](P<0.01),PTP1B表达水平(1 130.4±149.4)明显高于对照组(330.3±72.8)和APS干预组(485.9±85.6)(P<0.01)。结论 APS可改善肥胖小鼠精子顶体反应,其机制可能与其降低精子PTP1B有关。 Objective To explore the influence of astragalus polysaccharides (APS) on the expressions of protein tyrosine phosphatase 1B (PTPIB) and acrosome reaction (AR) in sperm from obese male mice. Methods Forty male C57BL/6J mice were randomly divided into control group (n= 10) fed with common diet and high-fat diet group (n= 30) fed with high-fat diet. Eight weeks later, 20 mice in high-fat diet group were successfully established obesity models and were subdivided into model group and APS group, with 10 mice in each group. Control group and model group received gavage with normal saline and APS group received gavage with APS 200 mg/(kg ·d). After 4 weeks, the expression of PTP1B in sperm was detected by Western blot, and sperm AR was measured by chlortetracycline assay in three groups. Results The body mass was significantly higher in high-fat diet group ((32. 8 ± 1. 7) g) than that in control group ((25.6±0.6) g) (P〈0.01). The AR rate was significantly lower in model group ((10.6±6.2)%) than that in control group group ( (44.7 ± 10.5) % ) and APS group ( ( 21.3 ± 8.5 ) % ) (P〈 0.01 ), and PTP1B expression was significantly higher in model group (1 130.4±149.4) than that in control group (330.3±72.8) and APS group (485.9±85.6) (P〈 0.01). Conclusion APS can improve the sperm AR of male obese mice, which might be partially involved in reduced PTPIB expression.
