摘要
目的:研究氯硝柳胺对人结肠癌SW480细胞生长、周期及凋亡的影响,并探讨其可能的机制。方法:CCK-8法检测氯硝柳胺对SW480细胞生长的抑制作用,流式细胞术检测氯硝柳胺对SW480细胞周期及凋亡的影响,Western Blot法检测氯硝柳胺对SW480细胞蛋白水平的调控。结果:氯硝柳胺对SW480细胞生长有显著抑制作用,其半抑制浓度为4.6μmol/L。与对照组相比,实验组SW480细胞G0/G1期比例增加,S期细胞比例减少,凋亡率增加(P<0.05)。实验组β-Catenin蛋白的表达较对照组下调(P<0.05),Cyclin D1表达无显著差异(P>0.05)。结论:氯硝柳胺可抑制结肠癌SW480细胞的生长,将其阻滞在G0/G1期,促进细胞凋亡,抑制Wnt/β-Catenin通路,是一个具有潜力的抗结肠癌药物。
Objective:To investigate the inhibitory effect of niclosamide on the growth,cell cycle and apoptosis of human colon cancer cell line SW480,and to explore its possible mechanism.Methods:CCK-8was performed to reveal the inhibition of niclosamide on the growth of SW480.Flow cytometry was used to determine the change of cell cycle and apoptosis after treated with niclosamide.Western blot assay was used to test the regulation of cell protein levels.Results:Niclosamide could inhibit the growth of SW480 cell,and its half inhibitory concentration was 4.6μmol/L.Niclosamide increased the percent of G0/G1 phase and decreased the percent of S phase in the cell cycle,and increased the cell apoptosis(P〈0.05).Compared with the control,Nicosamide decreased the expression ofβ-Catenin by 19.73%(P〈0.05),while the expression of Cyclin D1 showed no significant difference.Conclusion:Niclosamide inhibits the growth of colon cancer SW480 cells,blocks its cell cycle in G0/G1 phase,and inhibits the Wnt/β-Catenin signal pathway,so niclosamide is a potential anti-cancer drug.
