基于响应曲面法的芦荟大黄素制备工艺研究

摘要 采用响应面法优化糖苷酶催化芦荟苷生物转化制备芦荟大黄素的反应条件。在单因素的基础上选择反应时间、叔丁醇浓度、反应温度为变量,以芦荟大黄素的浓度为响应值,采用Box-Behnken试验设计原理进行响应面法分析,确定了酶法转化芦荟苷生成芦荟大黄素的最佳条件为:反应时间为4 h,V(叔丁醇)∶V(水)=3.5∶6.5,反应温度为43℃,芦荟大黄素转化率90.80%。 The response surface methodology was used to optimize the reaction conditions of biosynthesis and production of aloe-emodin with glucosidase as catalyst. Base on the single factors,reaction time,concentration of tert-butanol,reaction temperature were chosen as variables. The response surface methodology of Box-Behnken model was used to analyze the conditions of aloe-emodin's biosynthesis. As the result,the optimal conditons of Aleo-emodin's biosynthesis were determinze as following: the reaction time was 4 hours,the concentration 3. 5: 6. 5,the reaction temperature 43 ℃. Under those conditions,the highest conversion rate was 90. 80%.
