摘要 为了缩短牡丹无性繁殖周期,以凤丹白(Paeoniaostii‘Fengdanbai’)种子为材料,研究植物生长调节剂GA,、6-BA及根长对牡丹凤丹白成熟胚解除上胚轴休眠的影响,探讨能够快速解除上胚轴休眠的适宜植物生长调节剂配比和根长。植物生长调节剂试验结果表明,在未添加植物生长调节剂的对照中,上胚轴未萌动;添加不同浓度的植物生长调节剂后,上胚轴均能在2~5d内萌动,其中最快打破上胚轴休眠的处理是6-BA0.1mg/L+GA,1.0mg/L,而较快打破休眠且诱导出的胚苗生长状况最佳的处理为6-BA0.5mg/L+GA,1.0mg/L。根长度试验结果表明:不同根长度对解除上胚轴休眠有一定的影响,根长0~1、4~5on时打破上胚轴休眠所需时间最短,但以根长为3~4cm能较快打破上胚轴休眠且成苗率和真叶抽生率最高,成苗的各项形态指标较优,同时对胚苗可溶性糖、淀粉及可溶性蛋白含量的测定结果均验证了适宜的根长度为3~4cm。综上,当凤丹自离体成熟胚胚苗根长长至3~4cm时,使用1.0mg/LGA,和0.5mg/L6-BA同时处理,既可快速打破上胚轴休眠缩短成苗时间,又可收获品质较好的种苗。 The effects of exogenous hormone and root length on breaking of epicotyls dormancy of Paeonia ostii ' Fengdanbai' mature embryo were studied, so as to explore the suitable hormone ratio and root length for shorting the breeding cycle. The results showed that in the CK, the epicotyls didn' t sprout. After adding the different concentration of hormone, the epicotyls sprouted in 2--5 d, the fastest ratio of hor- mones which could break the dormancy was 6-BA of 0.1 mg/L + GAB of 1.0 mg/L. While the best treat- ment which could break the dormancy quickly and increase seedlings rate was 6-BA of 0.5 mg/L + GA3 of 1.0 mg/L. Different root length had a certain influence on epicotyls dormancy, When the root length were 0--1 cm,4--5 cm,the time of breaking epicotyls dormancy was the shortest. However when the root length was 3--4 era,the plant percent and true leaf extraction rate reached the highest level, meanwhile the breaking dormancy and the morphological indexes of plants seemed well. The test of soluble sugar, starch and protein content of seeding also verified that the proper root length was 3--4 cm. Overall,when the root length of Paeonia ostii ' Fengdanbai' mature embryo was 3--4 cm,treating with 1.0 mg/L GA3 + 0.5 mg/L 6-BA could quickly break the epicotyls dormancy, short the breeding cycle, and abtain sprout with the good quanlity.
