松郁安神方对失眠大鼠脑组织氨基酸类神经递质的影响 被引量:6

摘要 目的观察松郁安神方对氯苯丙氨酸(PCPA)致失眠大鼠脑组织谷氨酸(Glu)及γ-氨基丁酸(GABA)含量的影响,探讨其治疗失眠可能的作用机制。方法腹腔注射PCPA建立大鼠失眠模型,造模成功后,连续7d用松郁安神方进行灌胃。高效液相色谱法检测对照组、PCPA模型组及松郁安神方高、低剂量治疗组大鼠大脑皮层Glu和GABA含量。结果与对照组比较,模型组大鼠皮层Glu和GABA含量升高,Glu增加比较明显(P<0.001),且Glu/GABA比例增大(P<0.05);经松郁安神方干预后Glu均明显下降,与模型组比较,差异有统计学意义(P<0.01),且Glu/GABA比例显著减小(P<0.05)。结论松郁安神方具有镇静安神作用,其机制可能与调节兴奋性氨基酸神经递质的含量有关。 Objective To observe the effects of the compound Song Yu An Shen Fang on levels of glutamic acid ,γ‐ami‐nobutyric acid in cerebral cortex of insomnia rats .Methods The model of insomnia was established by intrapertioneal injection of PCPA .The rats were randomly divided into control group ,insomnia group ,and therapeutic group .All the therapeutic groups were treated with corresponding drug for one week .The levels of Glu ,GABA were determined by High Performance Liquid Chromatography .Results The levels of Glu in cerebral cortex of rats increased remarkably comparing with the control group (P&lt;0.001) ,and Glu/GABA proportions increased (P&lt;0.05) .However ,the levels of Glu of observation groups treated with different dosage of Song Yu An Shen Fang were significantly decreased (P&lt;0.01) and Glu/GABA proportions significantly re‐duced (P&lt;0.05) .Conclusion Song Yu An Shen Fang has the effect of tranquilizing and allaying excitement ,which is possible‐ly related to the excitatory amino acids such as Glu and GABA in the brain of rats .
