瘦素缺陷型ob/ob小鼠和C57BL/6J小鼠生物学特性的对比研究 被引量:6

摘要 目的对比瘦素基因遗传缺陷ob/ob小鼠与C57BL/6J野生型小鼠的生物学特征,为进一步的中药研究提供可靠的脂肪肝动物模型。方法将8周龄、雌性C57BL/6J野生型及ob/ob小鼠分别作为对照组和模型组,标准饲料喂养至16周。每周测量1次体质量和体温;检测血清肝功能(天门冬氨酸氨基转移酶、丙氨酸氨基转移酶)和血脂(总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白)含量;检测肝质量、肝组织中总胆固醇、甘油三酯含量,并进行肝组织HE染色。结果与C57BL/6J野生型小鼠比较,ob/ob小鼠体质量显著增加(P<0.000 1);肛温明显降低(P<0.001);血脂总胆固醇、高密度脂蛋白、肝重、肝脏指数、肝功能、肝组织中总胆固醇、甘油三酯含量明显增高(P均<0.01)。大体及组织病理结果显示,16周ob/ob小鼠肝脏表现为中、重度脂肪肝的病理特征。结论 ob/ob小鼠是较好的非酒精性脂肪性肝病动物模型,具有发病快、稳定、显著、重复性高等特点。 Objective To comparatively study the biological characteristics of leptin gene defect ob/ob mice and C57BL/6J wild-type mice, so as to provide a reliable fatty liver animal model in further study of traditional Chinese medicine. Methods Female C57BL/6J wild-type and ob/ob mice at the age of 8 weeks were selected as control and model animals. The mice were feed with standard diet for 16 weeks, the body weight and anal temper- ature of mice were determined every week, liver function and blood fat of serum were determined in the 16th week. The liver tissues were harvested for liver index, total cholesterol (TC), triglycerides (TG) content detection and HE staining. Results Compared with the wild-type mice, the body weight, liver weight, liver index, TC, HDL, AST, ALT of serum and TC, TG concentration of liver tissues of ob/ob mice were significantly increased (both P〈 0.01 ), and anal temperature of ob/ob mice were decreased obviously (P〈 0.001 ). General and histopathology showed that 16 weeks ofob/ ob mice performed the pathological features of moderate and severe fatty liver. Conclusion The ob/ob mice is proved to be a good animal model of nonalcoholic fatty liver disease, with the characteristics of fast, stable, significant and high repeatability.
