HIC1和SIRT1的异常表达与甲状腺乳头状癌的关系研究 被引量:4

摘要 目的探讨肿瘤高甲基化基因1(HIC1)和沉默信息调节因子2相关酶1(SIRT1)的异常表达与甲状腺乳头状癌发生、发展及转移的关系。方法利用免疫组化方法检测80例甲状腺乳头状癌组织及相应的80例癌旁正常组织中HIC1和SIRT1的表达水平。并根据免疫组化结果,分析甲状腺癌组织中HIC1及SIRT1的相关关系。结果 HIC1蛋白在甲状腺乳头状癌组织中的表达显著低于癌旁正常甲状腺组织(P<0.01);SIRT1蛋白在甲状腺乳头状癌组织中的表达显著高于癌旁正常甲状腺组织(P<0.01);HIC1与肿瘤的淋巴结转移状况、年龄及TNM分期相关(P<0.05);甲状腺乳头状癌组织中HIC1表达与SIRT1表达呈负相关(r=-0.759,P<0.01)。结论 HIC1和SIRT1的异常表达在甲状腺乳头状癌的发生发展中起重要作用,HIC1有望成为甲状腺乳头状癌预防和治疗的一个新的靶点。 Objective To investigate the expression of HIC1 and SIRT1 in papillary thyroid carcinoma and adjacent normal thyroid tissue, so as to explore its biological significance. Methods Immunohistochemical staining were used to analyze the expression differences of HIC1 and SIRT1 in tissues of papillary thyroid carcinoma ( 80 cases ) and adjacent normal thyroid ( 80 cases), and the data was statistically analyzed. Results Com- pared with adjacent normal thyroid tissue, the expression of HIC1 in papillary thyroid carcinoma was significantly lower (P 〈 0.01 ), while the ex- pression of SIRT1 in papillary thyroid carcinoma was significantly higher (P 〈 0.01 ).The expression of HIC1 was lower in the group with lymph node metastasis than in the group without lymph node metastasis (P 〈 0.05 ).Further clinical analysis demonstrated significant association of the expres- sion of HIC 1 with both age of patients and stage of disease (P 〈 0.05 ).The expression of HIC 1 and SIRT1 also showed a strong inverse correlation (r =-0.759, P〈 0.01 ). Conclusion Aberrant expression of HIC1 and SIRT1 in papillary thyroid carcinoma might play an important role in the on- cogenesis and progress of papillary thyroid carcinoma .HIC1 is expected to become a novel marker for prevention and treatment of papillary thyroid carcinoma.
