番茄红素对实验性糖尿病大鼠肾脏氧化应激损伤的保护作用 被引量:4

摘要 目的:研究番茄红素(Lycopene,LP)对实验性糖尿病大鼠肾脏氧化应激损伤的保护作用。方法:通过注射链脲佐菌素(STZ)破坏胰岛细胞的方法制备糖尿病大鼠模型,选取80只模型大鼠按血糖水平随机分为:糖尿病模型组、番茄红素5、10、20mg/kg治疗组和盐酸二甲双胍200mg/kg治疗组,并另取16只同龄大鼠作为正常对照组,每天灌胃给药一次,疗程4周。分别于给药前和给药后每周测定各组大鼠空腹血糖水平;治疗4周后,测定各组大鼠血清中血尿素氮(BUN)、血清肌酐(SCr)、尿酸(UA)含量及24h尿蛋白量(UPro),以及总抗氧化能力(T-AOC)水平和丙二醛(MDA)含量;测定各组大鼠肾脏组织匀浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量;通过HE染色观察肾脏组织病理形态学变化。结果:与糖尿病模型组相比,番茄红素20 mg/kg治疗组大鼠空腹血糖显著降低;番茄红素10、20 mg/kg治疗组大鼠血清中BUN、SCr含量显著降低,血清中和肾组织中MDA均显著降低,肾组织中SOD活性显著升高;番茄红素20 mg/kg治疗组大鼠血清中UA含量显著降低、TAOC水平显著升高,肾组织中GSH-Px活性显著升高;差异均具有统计学意义。番茄红素治疗组能够显著改善糖尿病大鼠肾组织病理形态学变化,其中以番茄红素20 mg/kg治疗组效果最为显著。结论:番茄红素对实验性糖尿病大鼠肾脏氧化应激损伤具有剂量依赖性的保护作用。 Objective: To investigate the protective effects of lycopene( LP) on oxidative stress in kidneys of diabetic rats. Methods 80 diabetic rat models made by intraperitoneal injecting STZ were randomly devided into five groups: diabetes model control group,LP 5,10,20 mg / kg treated groups and Metformin hydrochloride 200 mg / kg treated group,and selected another 16 same-aged rats as normal control group. The drug was given by intragastric administration for 4 weeks,once a day. Before and once week after the drug was given,the level of blood sugar was determined. Four weeks later,UPro and the content of BUN,SCr,UA,MDA and the level of T-AOC in serum were determined; the activity of SOD,GSH-Px and the content of MDA in the renal tissue were measured; and the renal tissue histopathological changes was observed by HE staining. Results: Compared with diabetic model group,the blood sugar level of LP 20 mg / kg treated group was significantly decreased( P〈0. 01); UPro and the content of BUN,SCr,UA in serum of LP 10,20 mg/kg treated groups was significantly decreased( P〈0. 05,P〈0. 01),the content of MDA in serum and renal tissue were also significantly decreased( P〈0. 05,P〈0. 01),the activity of SOD in renal tissue was significantly increased( P〈0. 05,P〈0. 01); the GSH-Px activity in renal tissue of LP 20 mg / kg treated group was significantly increased( P〈0. 05); and the renal tissue histopathological changes of LP treated groups were significantly improved,especially the LP 20 mg / kg treated group. Conclusion: LP had dose-dependent protective effects on oxidative stress in kidney of diabetic rats.
