摘要
目的:研究何首乌不同炮制品含药血清对H2O2诱导PC12细胞损伤的保护作用。方法:将何首乌生品、清蒸品、黑豆汁蒸品按20g/kg剂量小鼠灌胃给药,分别将其含药血清分别分成20%、10%、5%含药血清组,采用MTT法测定其对正常PC12细胞增殖的影响和对H2O2致PC12细胞损伤的保护作用,并用比色法测定SOD活性、LDH和MDA含量。结果:何首乌不同炮制品含药血清对正常PC12细胞增殖均无明显影响,含何首乌生品、黑豆汁蒸品各浓度血清和10%,5%含清蒸品血清可显著增强受损细胞内SOD活性(P〈0.05-0.01);10%,5%含生品血清、含黑豆汁蒸品各浓度血清和10%含清蒸品血清能明显降低受损细胞LDH释放量(P〈0.05-0.01);含清蒸品各浓度血清和20%,5%含黑豆汁蒸品血清能降低受损细胞的MDA释放量(P〈0.05),而含生品各浓度血清对细胞内MDA水平无显著降低作用。结论:含何首乌黑豆汁蒸品和清蒸品血清对H2O2致PC12细胞损伤有保护作用。
Objective: Research of serum containing different processed products of polygonum multiflorum Thunb on the protective effect of H2O2-induced PC12 cell injury were studied. Methods: 20 g / kg polygonum multiflorum Thunb,including raw product,steamed product and steamed product with black bean sauce were administrated intragastriclly. Serum of each group were divided to 20%,10% and 5% respectively. Proliferation of normal and H2O2-induced injury PC12 cells was detected by MTT. At the same time,the activity of SOD,the level of LDH and MDA in H2O2-induced PC12 cell injury were measured by colorimetric method. Results: Serum containing different processed products of polygonum multiflorum Thunb had no significant effect on proliferation of normal PC12 cell( P 〉0. 05). All serum containing raw product,steamed product with black bean sauce and 10%,5% serum containing steamed product could protect the H2O2-induced injury PC12 cells from potential oxidative damage by enhancing SOD activity( P〈0. 05 - 0. 01). 10% and 5% serum containing raw product,20%,10% and 5% serum containing steamed product with black bean sauce,10% serum containing steamed product could reduce the LDH level( P〈0. 05 - 0. 01). Each serum containing steamed product,20% and 5% serum containing steamed product with black bean sauce could depress the releasing of LDH( P〈0. 05). However,all serum containing raw product have no effect on the lowing of MDA level( P〉0. 05). Conclusion: The serum containing steamed product with black bean sauce and steamed product of Polygonum multiflorum Thun show protective effect on PC12 cell against H2O2 damage.
