鸡矢藤提取物对糖尿病小鼠血糖、糖耐量和胰岛素分泌的影响 被引量:3

The study of extracts of Paederia scandens on blood glucose,glucose tolerance and insulin secretion in diabetic mice

摘要 目的:研究鸡矢藤提取物对糖尿病模型小鼠血糖、糖耐量和血清胰岛的影响。方法:采用腹腔注射链脲佐菌素建立糖尿病小鼠模型,糖尿病模型小鼠随机分为链脲佐菌素对照组、二甲双胍组、鸡矢藤提取物2.5g、5g、10g/kg剂量组,另设10只正常小鼠作为空白对照组。各给药组小鼠灌胃给药每天一次,连续给药14天,空白组和链脲佐菌素对照组给予等量生理盐水。药前、药后7天和14天测定小鼠空腹血糖。末次给药1小时后进行糖耐量试验,测定小鼠血清中胰岛素的含量,并计算胰岛素敏感指数。采用苏木精-伊红染色法观察胰腺组织的病理变化,免疫组化染色观察胰腺组织胰岛素的表达情况。结果:与空白组比较,链脲佐菌素对照组的空腹血糖(26.58±4.53)明显较正常组高,血糖曲线下面积(115.30±8.35)增大,血清胰岛素浓度(13.19±4.24)明显下降。与链脲佐菌素对照组相比,鸡矢藤提取物10g/kg治疗组的空腹血糖(17.47±5.00)有不同程度的下降,血糖曲线下面积(84.86±9.56)减少,血清胰岛素浓度(18.29±3.77)明显升高,且鸡矢藤提取物也可以使胰腺组织中胰岛素表达显著增加,胰岛素敏感指数(-5.71±0.26)有所提升,但无统计学差异。结论:鸡矢藤提取物通过提高糖尿病小鼠的糖耐量,促进胰岛素释放,增加其胰岛素敏感指数,并改善胰腺组织的形态结构,使糖尿病小鼠空腹血糖下降。 Objective: To investigate the effect of extracts of Paederia scandens( EPS) on blood glucose,glucose tolerance and serum insulin in diabetic mice. Methods: Diabetic mice were created by intraperitoneal injecting STZ and randomly divided into STZ group,metformin group,low-,medium-,high-dose( 2. 5g,5g,10 g / kg) of EPS groups with 10 diabetic mice respectively. Normal group included 10 normal mice.Mice were intragastric gavaged once a day for 14 days. Normal group and STZ group were given saline. The fasting blood glucose( FBG) of0,7,14 days were measured. One hour after last gavage,oral glucose tolerance test( OGTT) and the measurement of serum insulin was conducted,the insulin sensitivity index( ISI) was calculated. Pathological changes of pancreas were observed through HE staining. Immunohistochemical analysis was used to perform insulin expression in the pancreatic tissues. Results: Compared to normal group,the FBG and the area under curve of blood glucose( AUC) were increased,serum insulin level were significantly decreased in STZ group. After the treatment of metformin and EPS,the FBG and AUC of mice was showed a reduction in varying degrees. While the serum insulin level was risen( P〈 0. 01). And EPS also could up-regulate the expression of insulin in pancreatic tissue. ISI was promoted by EPS,though the ISI result did not have statistically significant difference. Conclusion: The EPS can reduce FBG of diabetic mice through enhancing the glucose tolerance,promoting the release of insulin and raising ISI in STZ-induced diabetic mice.
