亲和体分子在影像、治疗和生物技术中的应用 被引量:1

摘要 亲和体(affibody)分子由58个氨基酸组成,蛋白相对分子质量约为6.5k Da,其来自于金黄色葡萄球菌蛋白A(SPA)B结构域。含有3个α螺旋结构,其中该亲和体分子的第1及第2螺旋中含有13个特定位点的氨基酸,并且这些氨基酸可随机突变,对其结构无明显影响,形成可以与任何分子结合的亲和体文库。亲和体的功能和抗体类似,但也有一些性质是抗体所没有的:如相对分子质量小、高亲和力、折叠速率快、理化性能稳定、能经受受化学修饰等特点,因此有人称其为"人工抗体",在治疗、诊断和生物技术中得到广泛应用。
