摘要
为有效控制贵州辣椒的病毒,采用生物学单斑分离法对酶联免疫吸附(ELISA)检测烟草花叶病毒为阳性的贵州辣椒病毒分离物(TMV-GZ)进行纯化。RT-PCR技术克隆其外壳蛋白(CP)基因并测序,使用DNAMAN7.0和MEGA5.0软件对TMV-GZ的CP基因进行序列同源性分析。结果表明:成功克隆该病毒分离物的CP基因,其序列长度为480bp,编码159个氨基酸残基;该分离物的CP基因与11种已报道的TMV各地分离物同源性均在98%以上,而与其他3种同属不同种的病毒同源性均低于65%。贵州辣椒感染的病毒为TMV。
The positive pepper virus isolate (TMV-GZ) isolated from tobacco mosaic virus by ELISA was purified to effectively control virus of pepper in Guizhou. The coat protein gene was cloned and sequenced by RT-PCR technique and the sequence homology of TMV-GZ^s CP gene was analyzed by using DNAMAN7.0 and MEGA5.0 software. Results:The sequence length and amino acid residues of the CP gene of the cloned virus isolate are 480 bp and 159 respectively. The homology between the isolate's CP gene and 11 reported TMV isolates is above 98% but the homology between he isolate's CP gene and other three viruses in the same genus is below 65%, which proves the isolated virus infecting Guizhou pepper is TMV.
