电针不同穴组对功能性便秘大鼠结肠c-kit蛋白及c-kit mRNA表达的影响 被引量:2

Effects of electro-acupuncture at different acupoints combination on expression of c-kit protein and c-kit mRNA in rat colon with functional constipation

摘要 目的:探讨电针不同穴组对功能性便秘(FC)模型大鼠的疗效,并检测其对大鼠模型结肠c-kit蛋白及c-kit mRNA表达的影响。方法:成年SD大鼠50只,随机分为正常组、模型组、电针1组(双侧天枢、大肠俞)、电针2组(双侧曲池、上巨虚)、电针3组(单侧天枢、大肠俞、曲池、上巨虚),每组10只。以0℃的0.9%氯化钠溶液灌胃复制FC大鼠模型,通过电针刺激干预10d,每天1次,每次20min,观察大鼠肠道炭末推进率、粪便含水量、c-kit蛋白和c-kit mRNA表达的变化。结果:与模型组比较,电针2组肠道炭末推进率、粪便含水量、c-kit蛋白及c-kit mRNA表达明显升高(P<0.01)。结论:电针可能是通过升高FC大鼠结肠中c-kit蛋白和c-kit mRNA的表达而改善结肠组织中Cajal间质细胞的形态和功能,使FC大鼠结肠蠕动功能得以增强,粪便形态得以改善,提示电针刺激曲池、上巨虚干预FC效果更好。 Objective: To observe the influence of electro-acupuncture at different acupoints combination on expression of c-kit protein and c-kit m RNA in rat colon with functional constipation(FC). Methods: Fifty SD rats were randomly divided into five groups: control group, model group, electro-acupuncture group 1 [bilateral Tianshu(ST25), Dachangshu(BL25) ], electro-acupuncture group 2 [bilateral Quchi(LI11), Shangjuxu(ST37) ], electro-acupuncture group 3 [unilateral Tianshu(ST25), Dachangshu(BL25), Quchi(LI11), Shangjuxu(ST37) ], ten rats for each group. Rats were given the frozen water by gavage to induce the FC model. The electro-acupuncture groups were treated by electro-acupuncture for ten days, once a day, 20 min for each time. Before and after treatment, the intestinal carbon propulsion rate, fecal moisture content, expression of c-kit protein and c-kit m RNA in colon were measured. Results: Compared with model group, the intestinal carbon propulsion rate, fecal moisture content, expression of c-kit protein and c-kit m RNA in colon were obviously increased in electro-acupuncture group 2(P〈0.01). Conclusion: The electro-acupuncture therapy could increase the expression of c-kit protein and c-kit m RNA in FC rat colon that contributed to improve the morphology and function of Cajal mesenchymal cells in colon, resulting that the colonic peristaltic function and the morphology of faeces were all improved. It suggested that electro-acupuncture at the acupoints of Quchi(LI11) and Shangjuxu(ST37) should have a better effect on functional constipation.
