摘要
目的:研究固本防哮饮对哮喘缓解期小鼠肺组织白介素4(IL-4)、白介素12(IL-12)的干预作用,探讨其治疗哮喘的免疫学机制。方法:75只小鼠随机分为正常组、模型组、预防组、固本防哮饮组和孟鲁司特钠组,以卵蛋白(OVA)和呼吸道合胞病毒(RSV)联合致敏激发制备哮喘缓解期小鼠模型。观察各组小鼠的一般行为活动和病理学变化特点,用RT-PCR法测定哮喘小鼠肺组织IL-4、IL-12的m RNA表达。结果:模型组小鼠肺组织可见支气管壁水肿、增厚,周围有嗜酸性粒细胞和中性粒细胞浸润。预防组、固本防哮饮组及孟鲁司特钠组,小鼠肺病理改变显著改善,支气管壁充血水肿现象减轻,管壁和平滑肌厚度减小,且嗜酸性粒细胞和中性粒细胞浸润也显著减少。模型组小鼠肺组织IL-4 m RNA较正常组显著高表达(P<0.05),而IL-12 m RNA较正常组低表达(P<0.05)。预防组、固本防哮饮组、孟鲁司特钠组IL-4 m RNA较模型组表达下降(P<0.05),而IL-12 m RNA较模型组表达升高(P<0.05)。结论:固本防哮饮防治哮喘的机制可能是通过降低IL-4 m RNA同时升高IL-12 m RNA表达来纠正Th1/Th2失衡,从而减轻气道炎性反应,缓解气道重塑。
Objective: By research the intervention effect of Guben Fangxiao Decoction on the expression of IL-4 and IL-12 in the lung tissue of mice with asthma in remission stage to explore the immunological and therapeutic mechanism. Methods: Seventy-five mice were randomly divided into normal control group, model group, prevention group, Guben Fangxiao Decoction group and montelukast sodium group. The asthma in remission stage model was established and induced with egg albumin(OVA) and respiratory syncytial virus(RSV). Meanwhile, changes of the mice in general activities and pathology were observed. In addition, Real time PCR was used to measure the levels of IL-4 m RNA and IL-12 m RNA in the lung tissue. Results: The lung tissue of mice in model group showed bronchial wall edema and thickening, which combined with infiltration of peripheral eosinophils and neutrophils. Nevertheless, the lung pathological changes in all treatment groups, including the bronchial wall edema and thickening and peripheral eosinophils and neutrophils infiltration, were all lighter than those in model control group.The level of IL-4 m RNA in model group was significantly higher than that in normal control group(P〈0.05), whereas the level of IL-12 m RNA in model group lower than that in normal control group(P〈0.05). The level of IL-4 m RNA in all treatment groups were all lower than that in model control group(P〈0.05), while the levels of IL-12 m RNA in all treatment group higher than that in model group(P〈0.05). Conclusion: The mechanism of Guben Fangxiao Decoction in the prevention and treatment of asthma might be closely related to the reduction of IL-4 m RNA and increased IL-12 m RNA that could correct the imbalance of Th1/Th2, so as to reduce the airway inflammation and alleviate airway remodeling.
