山核桃叶黄酮抑制TNF-α诱导的HUVECs与THP-1的黏附作用 被引量:5

摘要 目的:研究从山核桃叶中提取分离出来的黄酮单体2’,6’-二羟基-4’-甲氧基查耳酮对人脐静脉内皮细胞(HUVECs)与人单核细胞白血病细胞(THP-1)的黏附作用。方法:采用荧光标记检测经肿瘤坏死因子-α(TNF-α)刺激的HUVECs及THP-1之间黏附作用;采用荧光定量PCR及流式细胞技术,检测2’,6’-二羟基-4’-甲氧基查耳酮对HUVECs表面细胞黏附分子(VCAM-1)和细胞间黏附分子(ICAM-1)表达的影响。结果:2’,6’-二羟基-4’-甲氧基查耳酮能够明显减少TNF-α诱导的HUVECs与THP-1之间的黏附数量;能够抑制HUVECs表面黏附分子VCAM-1和ICAM-1的表达。结论:2’,6’-二羟基-4’-甲氧基查耳酮能够明显抑制TNF-α诱导的HUVECs与THP-1之间的黏附作用。降低HUVECs表面黏附因子VCAM-1、ICAM-1的表达可能是其抑制黏附作用的机制之一。 Objective: To study the influence of 2',6'-dihydroxy-4'-methoxy-chalcone ketone separated from the leaves of hickory on adhesive attraction of human monocytic leukemia cells(THP-1) and human umbilical vein endothelial cells(HUVECs). Methods: The adhesive attraction between HUVECs and THP-1 stimulated by TNF-α and THP-1 was detected by using fluorescent labeling. The influence of 2',6'-dihydroxy-4'-methoxy-chalcone ketone on expressions of intercellular adhesion molecule(ICAM-1) and vascular cell adhesion molecule(VCAM-1) on the surface of HUVECs were detected by using fluorescence quantitative PCR and flow cytometry techniques(FCM). Results: 2',6'-dihydroxy-4'-methoxy-chalcone ketone could reduce the number of adhesion between HUVECs and THP-1 induced by TNF-α significantly, and inhibit the expression of VCAM-1 and ICAM-1 on the surface of HUVECs. Conclusion: 2',6'-dihydroxy-4'-methoxy-chalcone ketone can inhibit the adhesive attraction between HUVECs and THP-1 induced by TNF-α and reduce the expression of VCAM-1 and ICAM-1 on the surface of HUVECs, which may be one of the mechanisms of adhesion inhibition.
