Aim:This study aimed to explore the potential anti-inflammatory effect and the underlying action mechanism of cucurbitacin E (CucE)by using phorbol 12,13-dibutyrate (PDB) +Ionomycin (Ion)-activated human peripheral blood mononuclear cells as a model. Methods:The expression levels of activated antigens CD69 and CD25 were evaluated by fluorescent staining together with flow cytometry. The pro-inflammatory cytokines interleukin-2 (IL-2),tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ)were evaluated by cytometric bead array. Results:The results showed that CucE significantly suppressed the expression of CD69 and CD25 on the surfaces of CD3 + T cells.Meanwhile,PDB +Ion-induced expression of IL-2,TNF-αand IFN-γwere also dose-dependently inhibited by CucE.Conclu-sion:These results demonstrated that CucE could inhibit the activation of PDB +Ion-stimulated CD3 + T lymphocytes in human PBMCs and decrease the pro-inflammatory cytokine expression, suggesting that it might exhibit anti-inflammatory activity via modulating the adaptive immunity.
