SIRT1在NMDA诱导的皮层神经元凋亡中的作用观察 被引量:1

摘要 目的观察沉默信息调节因子(silent information regulator 1,SIRT1)在NMDA诱导的大鼠皮层神经元凋亡中的作用。方法培养大鼠原代皮层神经元,给予NMDA(100μmol/L,2 h)建立兴奋性神经毒模型;应用Caspase-3活性分析和Annexin V染色方法,检测细胞凋亡。结果 SIRT1激活剂白藜芦醇(Resveratrol,RSV)拮抗NMDA引起的细胞凋亡,而SIRT1抑制剂Sirtinol(10μmol/L)取消RSV(25μmol/L)的保护作用。结论在NMDA诱导的细胞凋亡过程中,激活SIRT1发挥保护作用,而抑制SIRT1则参与神经损伤。
