补肾疗更浸膏对去势更年期模型大鼠下丘脑β-内啡肽与自由基代谢的影响 被引量:11

Effect of Busheng Liaogeng Extract on Free Radical Metabolism and β-endorphin in Hypothalamus of Ovariectomized Rat Model in Menopause

摘要 目的:研究补肾疗更浸膏对去势更年期模型大鼠下丘脑β-内啡肽含量与自由基代谢功能的影响,并探讨作用机制。方法:手术摘除雌性SD大鼠双侧卵巢复制更年期综合征模型,阴道细胞涂片检查模型成功后,将其随机分为正常组,假手术组,模型组,戊酸雌二醇片组(0.18 mg·kg-1),坤泰胶囊组(1.08 g·kg-1),补肾疗更浸膏低、中、高剂量组(4.5,9,18 g·kg-1),每组10只。除正常组、假手术组与模型组小鼠ig给予等量蒸馏水外,各组小鼠按照剂量要求ig给药,每天1次,连续4周。末次给药24 h后,腹主动脉取血,酶联免疫吸附试验(ELISA)法测定大鼠血清超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px)活性与丙二醛(MDA)含量,取下丘脑测定β-内啡肽(β-EP)含量。结果:与正常组比较,模型组大鼠血清SOD,GSH-Px活性明显降低(P<0.01),MDA含量明显升高(P<0.01),下丘脑β-EP含量明显降低(P<0.01)。与模型组比较,补肾疗更浸膏高、中剂量组大鼠血清SOD,GSH-Px活性明显升高(P<0.01),MDA含量明显降低(P<0.01,P<0.05),下丘脑β-EP含量明显升高(P<0.05);补肾疗更浸膏低剂量组大鼠血清GSH-Px活性明显升高(P<0.05)。结论:补肾疗更浸膏可通过调节去势大鼠下丘脑β-EP水平调控下丘脑-垂体-卵巢轴功能;并通过调节血液中SOD,GSH-Px活性及MDA含量增强去势大鼠抗氧化能力,起到延缓更年期氧化衰老进程的作用。 Objective: To study the effects of Busheng Liaogeng extract on β-endorphin( β-EP) in the hypothalamus and free radicals of rat model in menopause,and explore the mechanism of action. Method:Climacteric syndrome models were established by surgical removal of the bilateral ovaries of female SD rats. After successful modeling by vaginal smears test,the rats were randomly divided into normal group,sham operation group,model group,estradiol valerate tablets group( 0. 18 mg·kg- 1),Kuntai capsule group( 1. 08 g·kg- 1),Busheng Liaogeng extract low,medium and high dose groups( 4. 5,9,18 g·kg- 1),n = 10 in each group. The rats in various treatment groups received ig administration according to corresponding doses,once daily for 4weeks,and the rats in control group,sham operation group and model group were ig administered with the same volume of distilled water. 24 h after the last administration,abdominal aortic blood was taken to measure serum superoxide dismutase( SOD),glutathione peroxidase( GSH-Px) activity and malondialdehyde( MDA) content using enzyme-linked immunosorbant assay( ELISA) method,and hypothalamus was taken to measure β-EP content. Result: Compared with the normal group,serum SOD,and GSH-Px activity were significantly lower in model group( P〈0. 01),MDA content was significantly higher( P〈0. 01),and hypothalamus β-EP content was significantly lower( P〈0. 01). Compared with the model group,serum SOD and GSH-Px activity in Busheng Liaogeng extract high-dose and middle-dose group were significantly higher( P〈0. 01),MDA content was significantly lower( P〈0. 01,P〈0. 05),hypothalamus β-EP content was significantly higher( P〈0. 05),and serum GSH-Px in Busheng Liaogeng extract low dose group was significantly higher( P〈0. 05). Conclusion:Busheng Liaogeng extract can regulate hypothalamus-pituitary-ovarian axis functions by adjusting the level of hypothalamus β-EP in ovariectomized rats; and increase antioxidant capacity by adjusting SOD,MDA,and GSHPx levels in blood,with a delayed menopause role for oxidative aging process.
