褐参提取物对H_(20)荷瘤小鼠肿瘤抑制作用 被引量:1

Inhibitory effect of brown ginseng extraction on H_(20) tumor cells in mice

摘要 目的探讨褐参提取物对H20荷瘤小鼠的抑瘤作用及机制。方法 60只昆明雄性小鼠随机分为5组:模型组、环磷酰胺组、低、中、高剂量褐参组;H20造模24 h后灌胃给予受试物,1次/d、连续9 d,测量小鼠体重、瘤重、脾重、胸腺重量、白细胞数、血清白细胞介素2(IL-2)、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)水平及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果与模型组比较,高剂量褐参组小鼠瘤体重量[(1.37±0.42)g]明显下降,抑瘤率达39.38%(P<0.05);与模型组比较,高剂量褐参组小鼠血清中TNF-α、IL-2、IFN-γ水平[分别为(100.73±10.25)、(62.85±9.92)和(209.20±21.59)ng/L]及SOD、CAT、GSHPx酶活性[分别为(304.43±39.51)、(116.24±12.77)、(392.46±40.71)U/m L]均明显升高,差异有统计学意义(P<0.05)。结论褐参提取物对荷瘤小鼠肿瘤具有抑制作用,其机制可能与提高荷瘤小鼠免疫功能和抗氧化能力有关。 Objective To investigate tumor suppression effect and its mechanism of brown ginseng extraction in H20 tumor-bearing mice. Methods Sixty healthy male Kunming mice were randomly divided into 5 groups : saline group, cyclophosphamide group,low-, medium-, and high-dose brown ginseng treatment groups. Twenty-four hours after the establishment of H20 tumor model, the mice of treatment groups were gavaged with brown ginseng extraction of different doses one time a day consecutively for 9 days. At the end of the treatment, the weight of body, tumor, spleen, and thymus gland, the count of white blood cell ( WBC ), the content of tumor necrosis factor-α ( TNF-α), intedeukin-2 ( IL-2 ), and interferon-γ ( IFN-γ), the activity of superoxide dismutase ( SOD ), catalase ( CAT), and glutathione peroxidase ( GSH-Px ) were determined for all the mice. Results Compared with that of the saline group, the tumor weight ( 1.37 ±0.42 g) decreased significantly, with a tumor inhibitory rate of 39.38% in the high-dose brown ginseng group (P 〈 0. 05 ). Mean- while,significant increases in spleen index,WBC count,content of TNF-a( 100. 73 ± 10. 25 ng/L) ,IL-2 (62. 85 ± 9.92 ng/L) ,and IFN-γ(209. 20 ±21.59 ng/L) ,activity of SOD (304. 43 ± 39. 51 U/mL) ,CAT( 116. 24 ± 12. 77 U/mL), and GSH-Px (392. 46 ± 40. 71 U/mL)were observed in the mice of high-dose brown ginseng group. Conclusion Brown ginseng extraction has inhibitory effect on H20 tumor cells in mice and the effect may be related to the extraction-induced increases in immune function and antioxidation capacity of the mice.
