DHA对猪血液单核细胞炎症的影响

摘要 为研究二十二碳六烯酸(DHA)对猪血液单核细胞(PPBMCs)炎症反应的影响,本试验从猪外周血中分离PPBMCs,采用CCK-8法,观察不同浓度DHA(5、10、20μmol/L)对细胞活力的影响;采用荧光定量PCR和Elisa方法,观察不同浓度DHA(5μmol/L和10μmol/L)对PPBMCs中IL-6、TNF-α和MCP-1表达和分泌的影响;并检测不同浓度DHA(5μmol/L和10μmol/L)对LPS诱导的细胞炎症的影响。结果表明,5μmol/L和10μmol/L DHA对PPBMCs活力没有显著影响;DHA呈浓度依赖性地抑制PPBMCs中促炎因子(IL-6、TNF-α和MCP-1)的基础表达和分泌,并可抑制LPS诱导的促炎因子表达和分泌的增加。由此可见,DHA在一定浓度范围内可抑制PPBMCs的炎症反应。 In order to investigate the effects of DHA on inflammation,pig peripheral blood monocytes ceIIs(PPBMCs) was isolated from pig peripheral blood.CCK-8 was used to test the effects of different concentrations of DHA on cell viability;real-time PCR and Elisa was used to evaluate the effects of DHA on IL-6,TNF-α and MCP-1 or DHA on LPS induced inflammation.The resuhs showed that there was no effects of DHA at 5μmol/L and 10 μmol/L on PPBMCs viability;and DHA inhibited basal and LPS induced IL-6, TNF-α and MCP-1 expression and secretion in dose-dependent manner.In conclusion, DHA could inhibit the inflammatory effects of PPBMCs.
