HPLC-ELSD法测定酸枣仁胶囊中菝契皂苷元的含量

Determination of Sarsasapogenin in Wild Jujube Seed Capsules by HPLC-ELSD

目的：建立酸枣仁胶囊中菝契皂苷元的HPLC-ELSD含量测定方法。方法：采用反向高效液相色谱-蒸发光散射检测法。色谱柱为Waters Symmetry C18色谱柱（4.6mm×150mm,5μm）,流动相为甲醇-水（90∶10）,流速：1.0m L/min;柱温30℃;检测器漂移管温度85℃,载气流量1.8L/min。结果：菝契皂苷元在0.326-6.520μg范围内线性关系良好（相关系数为0.9996）。平均加样回收率为99.92%,RSD为2.06%（n=6）。结论：该方法操作简便,专属性强,重现性好,可用于酸枣仁胶囊中菝契皂苷元的含量测定。

Objective：To develop a HPLC-ELSD method to determine Sarsasapogenin in Wild Jujube Seed Capsules. Methods The HPLC method was performed on an Waters Symmetry C18column（4.6mm×150mm,5μm） with the mobile phase consisted of methanolwater（90∶10）,at the flow rate of 1.0m L·min-1;The temperature of the drift tube was 85℃ and the nebulizer nitrogen flow rate was 1.8L·min-1. Results The liner range of Sarsasapogenin was 0.326-6.520μg（r=0.9996）. The average recovery was 99.92%,RSD 2.25%（n=6）.Conclusion This method is simple,rapid,specialty,reproducible and suitable to test Sarsasapogenin in Wild Jujube Seed Capsules.
